Restarting mycelium from an old jar
    #28296399 - 04/26/23 03:54 PM (8 months, 28 days ago)

Hi Forum,

I am about to try to restart a culture that has been sitting in a jar for about 5 years I think ... it is originally a fully colonized quart jar of rye.

It has shrunk through the years and created a lot of yellow liquid but I am hoping that the insides of the lump will be ok.

My thought is that I will extract the lump from the jar and take pieces from deep within the lump and put onto agar in front of the flow hood.

What do you think about this?

Kindest regards
Johnny

How do you know the lump isn't just a bacterial blob that killed the mycelium sometime in the past 5 years? Open your jar while wearing a mask and gloves, make sure it isn't something dangerous.

Thank you for your response. My thinking is that I will transfer pieces onto agar and if I from this point on can observe mycelial growth rather than bacterial growth, that would indicate that is is ok.

However, I do become a bit worried when you seem to imply that it might be dangerous ... could it be dangerous somehow?

Kind regards
Johnny

Think about it. Five years is a long time. Imagine what could grow in an undisturbed jar in 5 years. Just about anything.

go for it, you should be using sterile procedure while working with agar anyways

Thank you guys, I will think about this a bit and then make a decision. If I go ahead with this, I will of course post an update describing my experiences.

Kindest regards
Johnny
