Hello and good afternoon fellow fungus fiends it is I, mushhead, I know it has been some time since we last had to roll our eyes at one of my posts. I have been off in dimension R-83 biding my time until I could return to bless you all with a hearty salutations.
I have recently gotten myself an induction style Presto 23 Quart Pressure Cooker

This thing is pretty awesome with how fast it heats up, given you have proper induction cooktops, you're off the races quick.
As always I got my grain ready with a wash, soak, and boil.
 Looks good!
 soaked in hot soapy water and polished nice and shiny I'll be using regular mouth quarts for this grow.
The spores I'll be using have been acquired form a reputable source and are to be assumed clean though syringes are never as axenic as we seem to think.

The cooktop: https://www.amazon.com/Secura-9100MC-Portable-Induction-Countertop/dp/B00GMCAM2G Not a bad little cooker at all, heats up the PC super quick, and does the job. I recommend something a wee bit better than this little guy but if you're strapped for cash it's a cheap option. The Pressure Cooker: https://www.amazon.com/Presto-01784-23-Quart-Induction-Compatible/dp/B07PYLD742 Drop this bad boy onto any almost any induction cooker that's at least 1800W and it wont do you wrong. The inside is just slightly wider than the original Presto we're all used to which makes for an easier time loading jars/bags.

 15psi was held for 90 min, I used about 3 Quarts of water, allowed for the water to boil before I sealed it and left it over night to cool for use the next day.
 Pulled my jars out this morning, tossed them into my SAB, and inoculated. During Inoculation I used a torch to heat the syringe needle after each jar to prevent infection. I used micropore tape to seal the vent holes, two layers is what I use, some people use one and it works just fine - I'm just paranoid. The gray blemishes on the jars is caused by calcium and other minerals left over by the water. You can get rid of this by using Vinegar which I forgot to add to my PC.

Germination and colonization is off to a good start. This is about 8 days, these jars sat mostly in darkness at ambient temps of 74f or 23c.

  Things are progressing as expected. B+ is a predictable cube and these jars will get shaken here in another day or so.

Alright It's time for a full update on all the jars. In total I've done 20 rye grain jars. This is spore syringe to grain, not recommended but I'm experienced with this process and it's just a cleaning method to gather prints for agar work. Finding the comtams before you spawn is what we're doing here, weeding out the possible time wasters. Here are the good looking jars, no irregular growth, discoloration, or other notable inconsistency that could perhaps be contamination.
Here we have the stragglers, again no unexpected or irregular growth, discoloration, etc. Just slow.

Then we have the suspects, these are jars with irregular growth, some discoloration, possible bacterial spots, etc.
 If anyone spots anything sus in any of my jars speak up, I'm no TC, only highly experienced and even the eyes of experience can miss things.

   Alas at long last, after some time, we have our fruits. And a surprised looking sprout, a plant that I'll never ID because removing it would mean destroying my mycelia, sorry little buddy.

   The little plant is looking alright. The mycelia in the last two pics is reacting to a bacteria that is present, this is perhaps why the plant is alive still. The battle between the bacteria and mycelia is providing the plant with food.

 Update on the larger tub, it's doing pretty well. I would get a better pic but it's on the bottom of a few tubs and I'm lazy.

  Here's some updated pics of the tubs. During this process I have reached my goal. One of the tubs has already given fruits and I have collected a clean spore print. I followed basic procedure to take a print, one print was done for 6hrs, the second for 24hrs. 6 Hour Print:
 The 6hr Print I really didn't care for so it got folded a bit funky and tossed into my emergency spore collection.
24 Hour Print:
 This is the print I used to make my plates.
I then made up Four spore plates.
 
Now we've reached a critical stage of this log. I have gotten my print, my plates look clean, and I now have a pretty clean culture. From here I intend to take transfers, propagate onto grain, and my next flushes with this culture will end this log. Stay tuned!

Alright! Another full tub update! Here are all the tubs from the jars that passed the contam check.(I say passed but what I really mean is "good enough") You will notice that some of them are having bacterial issues, though that does not matter to me. I am just cultivating this out to clean up the cultures by collecting spore for agar work as previously stated.
      
   You can see from the above that spore syringe to grain is a bad idea. However, since grain was at the time my only option to cultivate with, and because I did not have the funds to acquire additional equipment I worked with what I had. I expect within the next couple of days that these tubs mature out and I'll toss up pics of them fully fruited off. There are the exceptions:
  This is the only good tub out of the whole lot that looks at all healthy enough to keep around for a second flush.

Just a quick update on the one good tub out of the whole lot:
  After a little under ~20hrs and she's looking gorgeous as hell.
Additionally the other tubs are doing alright as well, except they're just slower due to the bacteria present, though I will say still quite as beautiful.

 Overall the harvest has been nice, dried weight at about 408g rounded down. Most of the fruits I've harvested have been fairly chunky with solid stipes and thick caps. During this time I have taken clones, spore prints, and will continue propagation.
  I did re-hydrate all my tubs, I allowed them to sit in distilled water until soaked(10hrs), then drained. I have already observed pins and an additional flush coming off most of them, will post pics soon.

Alright! So it's been a second since I posted an update. Here are the spore swab plates from this grow now. You'll notice wedges have been taken.
  
Here are the first transfer plates. You can see wedges have also been taken from these.
 
And here we have second transfers.
   
Soon these will be going to grain and the final culmination of this log will be those final tubs.
Here are the surviving test plates. The others will not be shown as they have been disposed of. 2% LME to 500ml water is your best bet with agar.

Alright so it's been a second since I've updated this and I want to say I've been pretty lazy with the cultures. I know that the plates are labeled GT, this does not stand for Golden Teacher, This stands for Row G, Column T. I store my plates with a categorized lettering system for easy retrial and identification so that I don't have to write all over my plates too much.
    
You can see that my agar work has been pretty good, there's some amazing looking growth there. I have given the cultures time to grown, mainly because I'm lazy and have had other cultures to work on but i wanted to finish out this thread. These plates look amazing. I'll be putting these to grain soon, tomorrow actually since today I am making more grain spawn fresh for the occasion. Can't wait for the coming tubs as I am quite confident we'll win out proper. Once my tubs have flushed out from this it will end this log.

Yep! to the left are about 30+ other jars, the five pictured, other than the one to the left are the ones I Spawned which were fully colonized.

Either the clones/ cultures are bunk or something is off with your suface conditions, i think its the latter,
Even with slightly bacterial sub You should be seeing much more even fuller pinsets, smaller tubs like shoeboxes were always harder for me to maintain, you might need thicker subs or just a little less fae. I
see a few that look good but most are on the dry side, one or two looks to have been sprayed/slightly matted but still on the dryish side,
I also find i get better pinsets when my top layer is thicker, but not super compressed. And semi controversial opinion, a straight verm casing is easy af and can dramatically inprove surface conditions in less than ideal climates, same with true casings, neither are necessary but can both make a huge difference in the right situation

I also find i get better pinsets when my top layer is thicker, but not super compressed. And semi controversial opinion, a straight verm casing is easy af and can dramatically inprove surface conditions in less than ideal climates, same with true casings, neither are necessary but can both make a huge difference in the right situation

Thanks for the advice! These tubs are sitting in a grow room that has constant air flow due to running a carbon filter within the room itself. The jars for these were bacterial(this was spore syringe to grain) and when pressed upon by my finger are fairly wet, although a couple were on the dry side this morning when I took the pics. The bacteria is slowing things down, no matter how hydrated the tubs are. I used a pseudo casing layer for each of these tubs and I'm really not wanting to use vermiculite anymore simply due to the amount of dust and debris it causes.

For sure, Maybe just try goin thicker with the too layer/pseudo. And bacterial spawn does go slower especially in wetter subs, I donāt compress it at all, spawn it on the dryer side and spray just the top layer after spawning to compensate and to avoid overdrying ⦠then check it and mist as knotting or bottom water when the pins are in.
Keep dialing in conditions and cultures, seems like you already know spores to grain is less than ideal so but faht makes it work, iām an agar guy but if i werent, iād rather make a small brf cake and transfer a pin clone to another and one more transfer before slurrying that to grain.
Another thought. The recirculating fan on carbon filter may be giving too much airflow but not providing enough fresh air, unless you also have an intake/exhaust setup too..

The room is set up with external ventilation so fresh air is constantly being cycled through the room itself. The filter is in place to capture scents from herbs growing nearby. I don't compress my substrates either, usually I try to leave them as fluffy as I can. I've been around for some time, I know faht makes spore syringe to grain work and I've done it multiple times before. It can be a serious shit show and I'm fairly lucky that it didn't turn into a big green mess this go around. I'm not afraid of a wee bit of bacteria. You perhaps maybe want to go back to the first page and read the whole log

Theres only one page and i read the whole thing, i just teread it to see if there was anything I missed, you didnt specify whether you compressed and your photo shows your top layer super thin, your pinsets are weak even for bacterial sub and im seeing alot of aborts that all point to me with issues with airflow. so i was merely offering suggestions about how I handle itā¦
But if you arent looking for input and more i interested in keeping a log of your grow then perhapsin the future a journal entry would be a better place for this as opposed to the open forum... Iāll see myself out

I appreciate your feed back and I was in no way being rude my friend. I took your advice to heart and I even admitted that some were dry as you had stated. I in no way wanted to make you feel as though you were wasting your time and I enjoy the feedback. If I wanted to a journal-log I would have done it that way to begin with <3
 EDIT: I also agreed with you that you should make your subs a bit fluffy, which is what I do, and indeed a thicker case would be preferable.

 Overall the harvest has been nice, dried weight at about 408g rounded down. Most of the fruits I've harvested have been fairly chunky with solid stipes and thick caps. During this time I have taken clones, spore prints, and will continue propagation.
  I did re-hydrate all my tubs, I allowed them to sit in distilled water until soaked(10hrs), then drained. I have already observed pins and an additional flush coming off most of them, will post pics soon.

Alright! So it's been a second since I posted an update. Here are the spore swab plates from this grow now. You'll notice wedges have been taken.
  
Here are the first transfer plates. You can see wedges have also been taken from these. Don't worry I saw the bacteria, it is the main reason why I transferred from that plate so early.
 
And here we have second transfers.
   
Soon these will be going to grain and the final culmination of this log will be those final tubs.
Here are the surviving test plates. The others will not be shown as they have been disposed of. 2% LME to 500ml water is your best bet with agar.

Alright so it's been a second since I've updated this and I want to say I've been pretty lazy with the cultures. I know that the plates are labeled GT, this does not stand for Golden Teacher, This stands for Row G, Column T. I store my plates with a categorized lettering system for easy retrial and identification so that I don't have to write all over my plates too much.
    
You can see that my agar work has been pretty good, there's some amazing looking growth there. I have given the cultures time to grow, mainly because I'm lazy and have had other cultures to work on but i wanted to finish out this thread. These plates look amazing. I'll be putting these to grain soon, tomorrow actually since today I am making more grain spawn fresh for the occasion. Can't wait for the coming tubs as I am quite confident we'll win out proper. Once my tubs have flushed out from this it will end this log.
