Contamination questions - does bacteria grow vertically in agar? And discolored agar? (images attached)
    #28178667 - 02/09/23 02:18 AM (1 year, 11 months ago)

I've been trying to clean up some samples, and on several agar-water plates I've managed to grow *something* white but it doesn't seem to be growing larger so I'm not sure if its some mold or what.. From the side it looks like whatever it is, is growing into the agar. Is this strong evidence to exclude the possibility of it being bacteria?

I also inoculated a few plates on MEA with activated charcoal, and noticed a weird brown color (more evident with lighting from the bottom). At first I thought it was metabolites from contamination, but then I realized that the "brown" areas had no more grey color. Also the donor piece of agar started off with charcoal as well, and over time the grey disappeared.

I also attached pictures of a plate that is definitely contaminated (with a big puffy white growth) to show how much charcoal is getting consumed, even where it looks like there is no growth...

Does anybody have experience with this phenomenon? Thanks!

With utmost care and precision,I work to achieve my fungal mission.
Sterilizing all my tools and space,to ensure not a single spore is out of place.
I carefully select the fungal strain and onto the agar, I make it rain.
With a steady hand and gentle touch, I inoculate the plate without a hitch.
Success is sweet when it's pure and true, and my agar plates are a testament too. A job well done, with no contamination, my fungi inoculation, a thing of  admiration.

Mycelium eats food colouring and it will yellow/brown depending on the colour used

Thanks! I definitely noticed that the mycelium eat blue food coloring but I was very surprised to see that it eats activated carbon. Mainly because I've seen people post pictures of fully colonized black agar plates that have no signs of yellow/brown while my plates are losing their black color even with minimally visible growth.
