Wood ear clone - first post :)
    #28129340 - 01/07/23 01:48 PM (1 year, 3 months ago)

Hi all!

I’m new here  excited for my first post! ( Noobish - recently got into mycology)
So I found some wood ear fungus at my local dog park on a wood stump it was growing awesomely and I yoinked some and put it in a paper bag to take it home have a go at cloning it.

Upon opening the paper bag some hours later I noticed that there was a lot of white powdery stuff on the paper bag when lifting the wood ear.

I took 4 samples from the wild wood ear;

1. A wedge of the wood ear (cleaned with isopropyl alcohol then halved it horizontally - took one side of the ear) flesh touching the agar and skin membrane facing upwards.

2. A swab of the white powdery stuff - which I assume is spores.

3. There was a really thick area at the base that was full jelly flesh - heated a scalpel red hot and cut out a cube piece.

4. A small flesh piece from the base of another cluster of wood ear (different from no. 3)

These are the results:
1.

2, 3, 4.

2. (White powdery spores?) 6 o’clock - bottom left
3. (Cube piece) 10 o’clock - top left
4. (Small flesh piece) 2 o’clock - top right

At first the spores (2) had most activity and the cube (3) and the small flesh (4) had a bit of contam and bacteria on it. The wedge had a little bit of contam as well but had a bit of good (I assume) activity happening. This was close to Christmas and New Years and I was away from observing it for at least a week and a half and came back and surprisingly saw that the cube (3) grew a massive ball of mycelium even though it had contam to begin with.
The wedge (1) was interesting as well it looked rooty.

I took transfers from the cube sample (3) and a clear area which looked like it had mycelium growth in the agar - not on the surface.

Some of the white mycelium from the cube (3) had contam / bacteria but more than half did not ( which is awesome ) and the agar that had the mycelium growth [near the centre]  ( I assume is mycelium growth ) grew quite differently - not sure if that is what I’m after or if it is just mould or bacteria contam - but it’s growing like mycelium.

QUESTIONS FOR YOU ALL WHO WOULD LIKE TO INPUT

Does anyone know what that growth is near the centre? Is that mycelium growth?
It seems to be growing at a quicker rate than the white mycelium.

The white mycelium is what I should be after right? It grew weirdly it and didn’t really spread thinly out - it grew out thickly compared to what I have seen on past posts on here and online.

Keen to know anyones thoughts on my clone attempt.

Cheers!
Alan

hello, I'm not familiar with this species mycelium but the growth near the center looks like mold to me
maybe someone else can provide more helpful info
welcome to the shroomery

edit: after searching old post I'm confused now

At first I thought it may be mould but it kind of grows like mycelium.
I was hoping someone familiar with this species may be able to shed some light on this situation. The sample as mentioned was taken from the same plate in an area that was clear of bacteria but looked like mycelium was growing throughout the agar below the surface level.

The mycelium ( white ) is growing rather interestingly as it’s staying dense as it spreads rather than branching out.

Here is an update -
I made a liquid culture from some of the mycelium from the agar plate and it turned out great

Inoculated grain from liquid culture;

It’s seems to have a darker appearance or has blackened the grain which is pretty cool - Here is a lions main jar I made at the same time you can see that it’s not as black

Any suggestions or tips for substrate mix for the wood ear?

as i mentioned before I have no experience with this species but afaik this is a woodlover so maybe some hwfp mix? what do you usually use for other woodlover species?

Just found a video and researchgate article

Biology and Cultivation of Black Ear Mushroom  – Auricularia spp
https://www.researchgate.net/publication/315727943_Biology_and_Cultivation_of_Black_Ear_Mushroom_-_Auricularia_spp

Cultivation in Compost
Raw materials

Sawdust -78%
Rice bran- 28%
White sugar-1%
Calcium carbonate-1%

Composting  of  sawdust  for  Auricularia cultivation usually  takes  five  days  to  produce  the  type  of  compost required.  Mix  thoroughly  the  weighed  raw  materials indicated above and maintain moisture level at 65-70%.

Pile the  material  in pyramid  shape,  turnings  should  give  at  2 days interval then supplements are added.

After completing compost preparation,  sterilized  and allowed  to  inoculation and  fruiting  body  formation  of  Auricularia

Rubber  saw  dust  when  used  as  a  substrate  for  the growth of Auricularia gave maximum yield of 180.67g in 3 harvests

The Falcataria.  moluccana is the most  suitable tree spp.  that  can  be used for making the substrate  for  cultivation

Composted substrates  supplemented  with  10%  wheat bran  supported higher yields in  any  substrates
