Who has Tissue culture experince?

I know there has to be someone on this wonderful forum who has dabbled in micro propagation.
I'm just getting my feet wet. The lack of knowledge is driving me crazy. I bought Cannabis Cloning Tissue Culture Media and plan on making a bunch of practice jars. From there I assume i need Cannabis Rooting Tissue Culture Media to bring them out of storage.

If there is anyone out there with knowledge please make your self known! I want to pour these jars but i'm unsure of how the jars should be sealed. Im totally interested in other species (like terrestrial orchids) but figured I would practice with MJ since its so forgiving.

I've been dabbling for over two years now!  It's actually what revived my interest in growing mushrooms.  As your aware, the set of equipment required for the two disciplines have a lot of overlap.

I got sucked in trying to micropropagate venus fly traps.  I have yet to go from explant to invitro and back.

That sounds right to me.  Usually the rooting tissue culture media has different ratios and concentrations of auxin(s):cytokinin(s)  I have yet to make it that far so no first hand advice to give.

Sealed tight is best.  Based on my reading and limited experience, plants invitro don't need much if any air exchange.  There are some exceptions, but limited.  I watched some videos by Plant Cell Technology and he was using polypropylene portion cups for running experiments or small batches.  Looked like the work great and they seal tight. I have some some phytojars and lids from phytotechlabs that I've been using. They are about the size of baby food jars, which by the way also work well if you can get the label goo off.

I've had limited success surface sterilizing and introducing explants into culture.  If you've never of endophytic fungi or bacterial it's a hard lesson.  Cultures can appear uncontaminated for weeks or months and then suddenly...  Kalanchoee is super easy and fast to micropropagate.  I'm currently running some practice jars of that.

Check out the kits section of phytotech labs.  There's a tremendous wealth of information in the kit documents.  Even if it's not a species that interests you, there's still gems of transferable information to be found.  The documentation is a bit hidden away... Pull up the kit, go to related documents, click on the info button and it will reveal the downloadable PDFs.

Also, dig around in the container section.  It will give you an idea of what is being used commercially.  Plastic deli containers are a common one!

Perhaps that's even where you got cloning kit?  They used to have a hemp multiplication kit, but it's no longer listed.

Thank you for the well thought out response its appreciated. Im gonna dig into the kit documents now.
Its weird that You tube videos are ALL like 7-8 years old. And they all say the same basic crap with no real info.

I did a little micro propagation at a cannabis grow 5-6 years ago. Definitely not a pro by any means but learned a lot in my short time there.

Our flow hood was a hand-me-down and didn't work great, I think that's why we had so many contaminations. Ended up just going back to regular clone cutting.

Like father fungi said, you want your containers sealed completely. No need for fresh air. Deli containers actually work really well and are cheap when you order in bulk.

One of the biggest issues/learning curves is adapting the plant to normal growing conditions once removed from the sealed media containers. They go from nearly 100 % humidity and zero air movement to usually a much lower humidity with air flow. Grow trays with domes that have vents will be your best friend!

Good luck with your experiments, it is a very fun and rewarding hobby! Especially when you start to do side by side comparisons with different auxin/cytokinin combinations. You'll find even different strains like certain combos more than others
