By the time you get to spawn to bulk, your spawn is either going to contam out or it wonβt. Problems are introduced well before S2B, and itβs too late to control for it. Everyone does their S2B and growing in open air, so it is definitively proven to not be a source of contamination.
If youβre experiencing high contamination rates, itβs happening further up the cultivation chain.

After sterilizing grain, it's good practice to leave jars inside SAB for a few days or a least leave a control jar that is not inoculated...

If not congrats you've sterilized successfully
Then its most likely a transfer from a dirty sample or need to re-evaluate your aseptic technique.

well Iβve limited the amount of cultivation work by buying ready made agar plates and grain jars, but otherwise yea no SAB at all, my approach may have been sub optimal

That's not sub optimal, it's guaranteed failure. All of your agar/spawn work needs to be done with a SAB or flow hood. Sterile work means sterile work. Make that SAB and look under the how to sections here on how to use it properly.

Lol. Iβve done several open air transfers and clones as well and have had 100% rate as well. But I still donβt recommend it. Thatβs just me being a lazy ass and not wanting to set up my SAB but needing to get shit done.

One was trying to culture Penicillium roqueforti from a piece of blue cheese, which I did successfully AFAIK without an actual microscope. I figure the chances of getting another random penicillium mono culture growing out of the streak pattern are pretty low.
The second was a clean up attempt of a Ps subtropicalis plate, which I had clean sister plates of anyway. Got away from the mold on the donor plate and avoided anything else floating in my kitchen.
Never actually done it with anything I was serious about keeping clean

Itβs bad practice of course but if Iβm harvesting and like a fruits characteristics Iβll usually just pop it to the side then take a quick clone from it in OA before putting it In dehydrator. My work space is a little limited and the SAB take up all the work area.

Since it is possible to store mycelium bits from LC or agar in distilled water at room temperature for extended periods of time, shouldn't it be possible to store pieces of mushroom or stone the same way? It would be like cloning but to distilled water.

Had a nasty germ plate i dgaf about. Did an open air xfer at my breakfast table. Got a few cleanish plates from it already and a backup swab so Iβm just gonna send it.

Thanks for the horrible inspo billy and others. I dont wanna setup a sab if i can make a quick one OA and get it back in front of the hood.
