Tidal wave agar
    #28152486 - 01/22/23 04:43 PM (1 year, 2 months ago)

Hey guys. I can't figure out if I'm making a post properly. It's been 1 week. Agar using spore print.  Grain jar with syringe.

Looks good. You should probably transfer atleast once before going to grain.

So take a section of a plate and put it on a new plate, then let that colonize. Do I need to be looking for anything in particular

Yes. I usually take what seems to be growing the fastest. Also away from satellite colonies.

Consistent, organized, radially symmetric growth is what you want.
Avoid anything especially thin, wet looking, scalloped, patchy, slow, discolored, etc.

It can be rhizomorphic (strand forming / ropey), or tomentose (fluffy).
Either one, or a mix of the two, is just fine.

Take transfers early if you are suspicious of contamination.
You can take them as soon as the myc jumps off the previous agar wedge or as soon as it starts germinating from spores if you want.
Typically, I like to wait until the growth is about the size of a quarter, but it is up to personal discretion.
Take your transfer from the leading edge of the mycelium, never further in if you can avoid it.

Take small transfers, even really small is fine.
The less source material the lower the chance of transferring something you don't want to the receiving plate.

It can be useful to label the transfer number on each plate. I label germination plates with T0 and subsequent plates T1, T2 ... etc.

You'll get an eye for what looks healthy and what looks dirty with time.
Healthy cubensis myc flows in a way contaminant myc does not.

Good luck!

Edit:
I like the "away from satellite colonies" advice from JW123 above too.

Yeah basically ignore what I said and refer to this well thought out and detailed response by Greg. Seriously much better than mine. People like him are what make this the best community for learning how to grow mushrooms. I am lazy.

4 days since first attempt at agar transfer from spore print.
