About 4 days ago I got the agar plates done. There was no contam in the LC but did see myc growth.

Oats were prepped with a rinse, clean, 30 min boil and dry, bagged in 2 quart bags and PC 3 hours after venting and hitting internal temp. After cooldown I open the bags in front of the flow hood, hit them heavy, 20cc into a 2 quart grain bag, and reseal.

Then it occurs to me 'if you roll it side to side you will spread it out and get more inoculation points', or so that was my theory. (which I had not tried before) So I roll shake the bags till I think it spread out well. Room temp was 73 - 75. Bags were sat on a shelf to wait.

Three days later I got no growth. Like, nothing. Which is unusual, typically I see at least a little white poking out of something at least.

I'll give it another day or so and re inoculate but, I have a theory. Maybe when I shook it, I spread the LC out far enough that it dried before it had a chance to dig into the grain. Maybe when you do a LC inoculation its important for the LC to have a pool of liquid to survive a couple days till it makes it into the inside of the grain.

Does this sound reasonable? Trying to explain why I got no growth from well prepped grain and verified good myc. I felt like I put in a lot of work and the shrooms just gave me the finger.

I dont know what is happening, but I do know that rolling your bag to spread the LC around is not the culprit.

Even microscopic portions of the LC are enough to start the process and all you need is one to survive and populate to colonize the bag.

20ccs for a 2 quart bag is excessive, and you might have waterlogged your grains making them less cooperative for colonizing.

Just wait another week or three and see how it progresses. While LC usually can be seen in a few days, there are factors (and too much water is one of them) that can slow it down. Good Luck!

Wait, to confirm, too much water in the bag can slow down colonization?

like days, or a week (?)

there's no way your LC dried out in the bags before it could colonize. like cooleko said, too much liquid can definitely slow things down & make it easier for bacteria to take off if it's present. 3 days isn't really that long imo & i've noticed slower recovery onto LC if i shake the grains vs just roll them around the jar without disturbing the grains much so both things coupled together probs compound. be patient

Yes. If too much water they might die before getting started or stall out for weeks. An excess of water slows mushrooms down and speeds competitors up, that is why we focus on keeping water levels optimal.

Since it was LC and hopefully clean you just have to be patient. If there were any contams floating in the lc syringe they will be pleased with the current conditions.

The bags were slightly more wet on the inside than normal. I didnt dry them well enough before bagging. Adding that much LC to that size bag wasnt helping. I didnt realise that could slow down colonization, I figured it didnt matter if they were still wet because of the length of the PC cycle (so not worried about contam) and bags do lose moisture over time through the filter patch.

I had read people prepping grain and wanting it to be completely dry on the outside before bagging but theres so much bro science out there that if I read someone's process, and there doesnt seem to be a logical explination for a process or ingredient, im not doing it.

I frequently go 'tim the toolman taylor' and P for plenty. I just figured more LC is 'more grow horsepower'. And waiting is for suckers so I'm not doing it.

Dry grain is important. Too much LC might be bad. Good to understand. Thanks for the help.

Did you test the LC on agar before using it? It sounds like you just looked at the LC to see if it is clean?

The statement was poorly worded. It was meant to indicate that the liquid culture had been applied to agar dishes and the only resulting growth was mycelial and not contaminated.
