Freeze Sterilization?

I have access to an extremely low temp freezer (in the -30C range) and I was wondering if I could just freeze the jars to sterilize them?

I think the glass of the jars are strong enough to withstand the low temps, but would it kill whatever nasties might be swarming through my substrate?

Need the mycology experts on this one.

your idea is preety good , i was thinking about this method too , but i'm not sure if the temperature could kill all the bad spores becose they are preety resistant and could maybe survive freezing ...

Im realy interested what will experts say , so guys , what do you think ?

im not claimin to be an expert, but heres my two cents. It would kill MOST of the baddies, but if you think about it i live in canada where we have up to -40C and in the spring they spores are all still here. So in my opinion it wouldnt be enough to do it. Give it a shot tho with a jar or two. Its worth a try

I don?t think it?ll kill any spores.

Intuition tells me that if I wanted to preserve my sporeprints forever, I?d keep them in your freezer.

Nope. It'd kill everything that was living, probably, but it wouldn't kill endospores, and might not affect normal fungal spores.

Low temperatures cause problems to living things primarily due to the destructive expansion of water when it freezes. Spores and endospores contain very little free water, so they can endure freezing.

what if you tried doing a reverse pasturization on it. warm up the jars like your incubationg them to allow the spores to germinate, then put them into the freezer. maybe repeat the process once or twice??? Just a thought though.

Submerge you jars in a water bath for 24 hrs prior to your freezing, this will cause endospores to germinate, making them suceptible to temp extremes. I soak my jars prior to pcing anyway, cause although we soak the grains for the same reason, it doesn't mean that there aint any in the jars, especially if used before.

low temps like that will NOT kill endospores...

I think I'm going to incubate for a good 5 days to a week. This way, when I freeze em, it will kill the lively, and the spores should be long gone and germinated.

Just do a normal fractional sterilization by steaming instead of freezing.

There's a reason that medical facilities use autoclaves, and not big fucking freezers.  :smile:
