Clear agar plates but still liquid, what did I do wrong?
    #28017560 - 10/26/22 12:54 PM (2 years, 2 months ago)

So I made a set of plates and I really wanted to try to make them clear, here's what I did,

I had 2 quart jars filled to 600ml of water.
I added 12 grams of agar to them
7 grams of LME.
I stirred both up while cold, heated both separately in a pot for about 10 minutes while stirring.
I strained both of them with some cheese cloth.
Then put both back in the quart jars.
I pressure cooked at 15psi for 30 minutes.
One of them turned out alright so far. Haven't been tested yet.
The other is incredibly watery.
But as stated, I haven't tested the more "solid" one yet.
It looks to be pretty solid.
Any ideas what went wrong?

I would guess the filtering. LMEA does not need any filtering, it is going to be already very clear. I only noticed it cloudy when I add nutritional yeast to the LMEA.

Just give it a try without filtering. I think that should do the trick.

Edit: Cloudy Agar isn't an issue either.

So you dissolved your agar and malt extract, and then filtered the solution afterwards?

If so, you may have filtered out a bunch of the agar accidentally. If you're dead-set on clear, filtered agar plates, I would recommend dissolving your malt extract, filtering, and then dissolving your agar in the solution afterwards.

Yes I did exactly that. I noticed while filtering that some of the agar was coming out. Unfortunately I believed that's what did it. Wasn't sure though.

I think that sounds like the best solution. I was going to try this tomorrow morning.

The only reason I want such clear agar is because, I've noticed some molds or contam that I've gotten I've been able to spot a lot earlier and better from the bottom under all the myc, rather then the top. Especially since I've been struggling with temp control and condensation on some lids.

I completely understand. When making agar, I filter my nutrients as well. Add the agar after the filtering step and you're golden!

Yeah, agar only disolves at temps above 90C so gently heating it in jars and then filtering is going to take a bunch of it out.
