Have any of you made a sterile spore print straight to agar? Let an agar plate fruit, cut the cap off and place directly on another plate to print directly onto the agar?

Why not just cut some of the first and place it on the 2nd, the spores are just doing the same exact thing over again. The only reason too do something like you are saying is too get 100% clean spore print then stop. you would be going back a step, not moving forward.

Sorry, I know jack shit about this process so I may just be using incorrect terminology.
I thought that you were talking about cloning from one plate to another instead of the hypothetical tiny spore print. My understanding is that cloning/mycelium plate transfers can’t be done indefinitely due to genetics degrading, but selecting fruits and using spores can be done forever.

You can make pretty clean spore prints by following teks on here. I wouldnt bother with making a tiny spore print from a fruiting agar dish.

I’m not too familiar with anyone cloning clones to the point of degradation, not that it isn’t possible. Lots of people have come up with good cultures from cloning clones of clones, sometimes it doesn’t go as well like I have a clone that makes a weaker cake than the MS it came from but that’s got more to do with luck of the draw. And to answer your first question. Yes, if your culture fruits on agar and produces fertile spores you can swab/streak, those to another plate, if you put the cap to “print” on the second plate it would more likely clone to it faster than the spores would germinate. Transferring pins from a plate are a great way to transfer. Lots of people do that.
