Should a noob use a loop?

Just wondering what most would think is best? For a total noob going syringe to agar. Skip the loop and just put a drop in the center of the plate,  or does a loop help? Just not sure if it would be beneficial or just another way to make a mistake? Im assuming you would also flame the loop prior to use, and between uses?

Just drop a little dot in the middle wait for some growth and transfer. no need for a loop for what you are doing imo

The loop helps separate cultures, which can make choosing transfers easier. If you’ve got a huge mass of matted mycelium and you’re trying to pick the most “robust” growth to transfer, it may be hard to see what is what. If you’re just going from MSS to agar to be sure you don’t have contams, a drop or two on the plate should be fine. That being said, a zig zag with a flame sterilized loop is very fast and easy. If you can spare a plate, try both methods and see for yourself which you like best.

It's highly recommended to use a loop. Read BOD's DIY Inoculation Loop TEK.

You need to drop from syringe a single drop into inoculation loop. The trick is to choose a inoculation loop with the tiny loop and flexible enough not to cut through agar. The tiny loop only allows an exact amount of water, so adding more is just going to spill over, but the remaining water is always in the same quantity

You could use an advanced plate streaking technique is described in c10's Agar Guide.

The main idea is that you want to randomly distribute the spores and contaminants in colonies, and in some of them, your spores would dominate, having a better chance to fight.

A good video for plate streaking technique https://www.youtube.com/embed/_s5Fwgk_k1o. Because each streak section is created out of a previous section, size of colonies is getting smaller.

I prefer the above plate streaking technique to zigzag pattern because gives you a greater chance to find a healthy colony. The disadvantage is that you need a bigger plate to work comfortably.

I recommend that you use a loupe and drop a single drop on a plate. Try both methods there locally and observe. Teach yourself something and then you know how you prefer to work with syringes on agar

For me personally it does not make a huge difference whether the spores are plated really well or not, usually I go with a drop when it's a syringe and use the loupe if working from a print

Both methods are usually satisfactory for me here

I would argue using a loop later would be a better idea. Loops are sorta an advanced tool. I highly recommend getting a pair of medical tweezers. They work great when working with colonized agar.

I would recommend a BRF puck, then transfer mycelium to agar.

I struggled getting syringes to germinate on agar before I started using pucks..

The advantage of dropping on a loop and streak versus dropping on a plate and streak is that you have better control over the quantity of water dropped on a plate. It doesn't mather if you are really bad with a syringe or have shaking hands.

It takes a lot of practice to be able to drop a single drop on a plate from a syringe. Most of the time you'll create a pool on your plate and that's bad news.

So you are dropping the drip on the loop, and swiping the loop on the agar? It seems like it would be difficult to drop a single drip on the loop. I will be starting soon, so this thread is helpful.

It doesn't mather how much you drop on the loop. Only a single drop will remain on the loop's eye. Water on loop's eye forms a sphere at equilibrium.

Use a loop with a tiny eye to reduce the remaining amount of water in loop's eye.
