P. Mexicana Colonization Time Experiment Using Differing Agar Wedge Sizes with Oats

Hello!  Some people were telling me that inoculating 12 quart jars per agar plate would be too slow and that I should consider using much larger slices. Since I had some extra jars and extra plates, I decided to do a bit of an informal experiment. 

As I stated previously, I used 1/12th of a 100mmm agar plate to inoculate 12 jars.  I then inoculated 12 other jars using 1/6th-size wedges taken from two more agar plates.  I shook each jar vigorously after inoculation in my SAB, and I made sure that each wedge didn't stick to the roof of the jar lid.

I plan on doing routine updates since there is some debate about how large/small of an agar wedge should be used to inoculate a grain jar.

Grain: Oats
Petri Size: 100mm
Agar media: Potato water/Karo/Agar
Species: P. Mexicana
Strain: Galindoi
Isolate?: No

Quick update at day 4.  All jars look to be in good shape.  I haven't shaken any, yet.  I'll probably wait until day 7 or later to do that.  The 1/6th wedge is on the left, and the 1/12th wedge is on the right.

Nothing really to update at this point.  Most jars look like the two above.  I'll probably let them colonize for a few more days before shaking. I'll update at that point.

I shook all of my jars today.  Most of the 1/6th wedge jars look a little more colonized than the 1/12th jars, but not by a whole lot. I'll update once they start recovering.

In total, I found about 4 contaminated jars among the two dozen I inspected today.
