Swabs to agar

If you had 6 swabs gifted to you months back, 3 varieties, and you wanted to make agar plates today and swab them with your swabs, how many would you make?
Can you make 20 plates with that?
Assume you tried pulling off a old print and got nothing out of 20 plates last attempt

So many numbers lol I suppose seems how it's 3 varieties I'd swab 3 plates, save 3 swabs to try incase they dont take.

Here is an easy way to do 2 plates per swab:
Cut the tip off. Stuff it in a plate. Use the rest to swab another plate
(I would also use only 3 like Rumfor said and save 3 for later in case - that magician is a wise one)

I'd start 3 plates of each variety, giving you 9 starts.

So like take one of each swab, cut out a bit and plant it in a plate, swab another plate with the leftover, make 6-9 plates from 3 swabs total, save 3 swabs for later ?

Steril water can help for sure. I'm lazy so I do that as a last resort with the backup swabs lol
You can always just suran wrap extra plates and save for later like ya say. You'll need em for transfers anyhow with all the success you're about to have!

I use to do 10g dme and 10g agar in 500ml water, will that work or do you need to alter your ratio for any reason?

It'll work.
Most people use LME and not DME.
Some people like softer plates for germination (maybe 7g agar/500ml) - I don't care about the softer plates as I use a loop and it does deposit water on my agar

You don't even need to look it up.
Strain your grain water after cooking the grain.
Strain again through a coffee filter. Mix 500ml GW with 9g agar (or whatever quantity is your favorite) and PC for 15-30 min - I do 15 but most people here seem to think 30 is safer.

I have some prints too it looks like, I'll try hydrating them with some of this water and transfer to some agar and see if I can get any of those going

Hydrate your swabs before swiping them. For a print, use a loop or a flame sterile knife to scrape some spore print.

Am I alone doing it this way?

Last time I tried to use prints I just scraped the print onto agar.
I made 20 plates with growth on 2 that didn't look promising, didn't even use it
I was planning to open the foil print, put water straight into the foil, mix around, then swab to some plates

That's definitely not how I do it. Flame sterilize metal loop, let it cool, swipe spore print then streak plate. No Water used. Water would just be a contam vector IMO.

I sterilize the water in a very small condiment cup. Seems to work just fine for me.
That being said, I will def give it a try without the water next time to avoid the extra step that suddenly doesn't feel necessary anymore

Guess we'll find out how it works out
Maybe next weekend I'll do the same thing and leave out the water and use up another swab and print

Dont worry about rhizo growth. It just needs to be clean, uniform, and organized. Often times germ plates won't throw rhizos till you transfer a couple times.

Impossible to tell from those pics. I see uniform shapes and that's good at least. Typically you wanna transfer from germ plates fairly quickly before contams culture there too and take over if present. Basically if you can grab a rice sized piece that's had a chance to get away a little bit from your inoculation point then you should do so.

I wanted to update this
So I ended up making transfers from those plates

Growth all looked clean so I took them to grain, this is some of the growth on the second transfers

And this is what the grain looks like now, innoced on 10/30, shook two days ago

So it seems like using the water worked alright for me
Last time I attempted to put spores to agar it failed and I didn't use water. Idr if I used swabs or exclusively prints or if the prints were dif
