Wait what?
When using a loop, I scrape the spores in a small amount of water, dip the loop in that water and then streak plates.
Am I alone doing it this way?

Last time I tried to use prints I just scraped the print onto agar.
I made 20 plates with growth on 2 that didn't look promising, didn't even use it
I was planning to open the foil print, put water straight into the foil, mix around, then swab to some plates

That's definitely not how I do it. Flame sterilize metal loop, let it cool, swipe spore print then streak plate. No Water used. Water would just be a contam vector IMO.

I sterilize the water in a very small condiment cup. Seems to work just fine for me.
That being said, I will def give it a try without the water next time to avoid the extra step that suddenly doesn't feel necessary anymore

I just hydrated everything bro and put it to plates
Guess we'll find out how it works out
Maybe next weekend I'll do the same thing and leave out the water and use up another swab and print

Dont worry about rhizo growth. It just needs to be clean, uniform, and organized. Often times germ plates won't throw rhizos till you transfer a couple times.

Impossible to tell from those pics. I see uniform shapes and that's good at least. Typically you wanna transfer from germ plates fairly quickly before contams culture there too and take over if present. Basically if you can grab a rice sized piece that's had a chance to get away a little bit from your inoculation point then you should do so.

I wanted to update this
So I ended up making transfers from those plates

Growth all looked clean so I took them to grain, this is some of the growth on the second transfers

And this is what the grain looks like now, innoced on 10/30, shook two days ago

So it seems like using the water worked alright for me
Last time I attempted to put spores to agar it failed and I didn't use water. Idr if I used swabs or exclusively prints or if the prints were dif
