My first run. Advice needed.

Hello. I just started my first attempt at a grow. This my my first spore to agar from inside my SIB using a swab.
 I also did a couple of all-in-one bags with about 4 cc's each from the same spore syringe flame sterilized between each use. I also did one bag of Uncle Ben's.
So, I am under the impression I should do spore to agar first. If this is best, why? When I inoculate my bags, guides say a dark area. Can I do this with a sealed storage container? I have yet to get past colonization of grain yet, but when I need to go to bulk, how do I decide what substrate to use? I have seen 90s on YouTube and some others use coco-coir, and some use cvg. IF I am growing z-strain (a manure loving strain), do I have to use a manure based substrate? Any help will be appreciated and I will update with more pics as I go.

Spores to agar is best because it lets you clean up the resulting colonies by performing subsequent transfer. Spores to grain is not ideal, as explained here. Next, cubensis benefits from indirect lighting at all life stages. No, cubensis do not need manure, and coir or coir and vermiculite is beat. Regretfully, your plate appears heavily bacterial.

That agar plate is straight bacteria. If you put that to a bag you're in for a bad time.. your best bet would be to make some brf pucks in half pint jars or no-pour containers, 2 parts vermiculite, 1 part brown rice flour and 1 part water. Steam or PC them and squirt some solution from your syringe in there. Brf is more resistant to bacteria, and your spores will have a better chance of germinating and outrunning the bacteria. Then you can transfer from there to agar and clean it up further.

Ummm.. That is not good. Hopefully your syringe is cleaner. Place 1 drop on center of new plate as that swab dosent appear salvageable.

IF I am growing z-strain (a manure loving strain), do I have to use a manure based substrate? coir, or verm and coir Any help will be appreciated and I will update with more pics as I go. good on you for jumping in with so many teks, thats how you learn fast. unfortunately you are off to some bumpy starts. i recommend starting with PF TEK while also playing with agar on the side. and remember to PREP EVERYTHING YOURSELF if you want results!

I bought the syringe off (advised to remove vendor) and other stuff off amazon. This is actually my second plate with the same results. I got kids science plates from amazon. I am waiting on some new agar plates from Mush Cult on etsy. I also got a new syringe from (advised to remove vendor2), so we will see how it goes. If I dont get good results, I will prep my own agar and select a new spore vendor. Im also looking at sourcing mats for PF tek. I will post a few pics from my bags, and plates when they come and things get going. Thanks all for the help.

and second on BRF pucks. Also, IMO those reddit vendors are whack and should be avoided. That said, hereโs an oldie but a goodie on prepping your own agar. For pre-pours, I use small widemouth mason jars (either 125 or 250 mL) with unmodified PP5 lids, and I leave them slightly loose during the PC cycle to avoid the extra steps of making GE holes, using micropore tape, foil, etc. Good luck, my tasty blueberry.

 These 2 bags are from the old spore syringe. Going to attempt to fruit in a different area soon.
 These are a new spore vendor and bag vendor
 First attempt at PF tek
 I do have condensation inside the jars. I'm thinking maybe it has to do with the Ambien temperature of the room? A baby, not sure why?
 New spore vendor and agar dishes. I'm not really sure where I should pull transfers from I already cleaned the contamination off the sparse one.
Any assistance is greatly appreciated. Thanks to all for the advice.
