Hey all, fiddle here.
Wanted to check in with everyone and share a grow-in-progress.
First I'm sorry for the length of this post.

I debated at length about whether to post this or not. I'm shy and awkward usually, even online. Part of me wanted to wait until I had pictures of harvests to share with the community. Something substantial. Another part got really giddy and couldn't wait to share. It won.

I feel like all the knowledge I've gained on here I want to give back any way I can.
To anyone who takes the time to read this, thank you. It is my hope that any errors or rookie mistakes I've made will be clarified, and that something in this TL;DR will help someone else out. To anyone who takes the time to read this, thank you.
For this grow I will be using a 16qt Presto PC and SAB. Agar wedges will go to spawn, and I will then put to CV in dialed MP monos.

note: I used oats with this grow. After further research, I do not recommend oats,  even though I haven't run into any problems yet. I am not advising you use oats or even recommending it. But it's what I did. I will most likely use millet or WBS next)

Edit 9-19-22

Prepped oats today.
No soak, rinsed twice and boiled on high (rolling boil) for 40 min.
A few burst hulls. Good.
I chose to take the risk following boil & strain and waited 2hrs, until grains appeared completely dry. (no shiny on the hulls)

"I personally find that drier grains perform way better than anything too wet. I sometimes prepare lots of grain a day ahead of time let it dry in the colander for a few hours then put it in the fridge overnight. It almost seems too dry, you can't get it too dry. If I have the time I vastly prefer to make the grain on the dry side."

Grains packed into 14 1qt jars with plastic lids 2/3 full ea. Plastic lids adjusted to app. 3/4' turn from finger-tight. (see above)  Grains sterilized @15psi for 2hrs in 2 rounds.

(note: 15 cups of dry grain makes just enough for this recipe, and can be done in one boil)
(subnote: I do not recommend over-filling the pot like I did. Although I did it successfully, you should follow the ratios listed in your chosen tek.)

edit: Fuck, I didn't give the jars long enough to cool before tightening the lid after cycle. Created a fucking vacuum. Not shmeated.

Here they are after the cycle. My 16qt Presto holds 7 jars. I would prefer to have a 23qt, but this is the only one I have access too. I miss my 25qt All-American. :/

Edt: Didn't remember to vent the PC before it was too late. Rookie mistake. Always remember to purge your autoclave, boys and girls. Not doing so is HIGHLY UNADVISED. (9/22/22)



Edit 9-20-22

It's that time again...



I decided to use some robust G3 TKS and B+ transfer petris to inoculate my grain spawn. Inoculation took about 45min from start to finish. I wanted to use Pastywhytes' tic-tac-toe Tiger Drop, but had to adapt it for disposable petris rather than the glad mini-rounds Pasty uses. The sterile procedure outlined in the tek, however, I followed to the letter.

As is pictured below, the agar plates were fully colonized, past-fully, really. I had them in the fridge for 8~ days prior.
Here is a brief breakdown of the transfers from what I could decipher from 8 pages of fine-written notes. I regret not documenting this more clearly and professionally, as well as the lack of pictures.

The entire lineage is descended from a single spore print I took from some KSSS and B+ I grew on pf cakes. An initial round of PDA pastyplates (8~) were inoculated with KSSS with a loop from print. At this time I also inoculated (10~) petri plates of B+, using an outdated and risky agar method. However I was able to take several donors and clean up sectoring a bit over 3-4 "gens."

I did the same with the TKS in petris over roughly the same amount of transfers. Performed with an all metal craft knife and solid metal tweezers, and a WhipIt brand butane torch.



As you can see, some of the cultures are tomentose, some more clearly defined growth.



I cut a line down the middle of each petri dish, making two small perpendicular scores across the surface of the plate, like this:



I picked up each half of the wedge using the scalpel as a skewer (after liberating PDA from the bottom of the petri, being careful not to let any part of the scalpel other than the blade to hover over the jar. Plastic lids tightened before shaking and then loosened to 1/4' away from finger tight. The entire process took about 45min start to finish.
~Notes to come later on which exact plate went into what jar. TKS #8 (transfer 4) was split between jars 1 & 2.

On tightening the lids:

Pastywhyte said "Step 3. Loosen 1/4" to a 3/4" turn from finger tight. This keeps the pressure from messing up the lid. You will keep the lid loose like this after inoculation as well. This will allow for plenty of GE for colonization."

I struggled a bit to find the perfect degree of twist to allow for optimal FAE during colonization. I ended up settling just shy of 3/4" I stopped the second I felt even the slightest contact with the threads. I felt like this was really important. I tried 1/2" first (as well as I could gauge) and when I checked back on em they seemed just a molecule too tight. (see below) It is a possibility I had the lids threaded just a fraction too snug on the jars lagging behind, preventing "optimal" FAE during the first 2-3 days of colonization. Caution: I am an over-thinker. Note to self: only touch jars by glass sides when handling, so an not to disturb lid.

Here they are post-inoculation.





Hoping for the best here but not counting my chickens. I predict a 20% failure rate on these (my average over the last 2mo.)

Checked on my jars about 24hrs later and they are showing signs of healthy growth. So happy.





Update 9-22-22

Jars are looking fabulous. Explosive growth at the noc points and plenty of colonized satellite grains already at only 48hrs. The lids were a little tight when I checked 'em, so I loosened them up about another 1/4' (very carefully) being conscious not to let my hand slip and unscrew too much.

Update 9-25-22

Some of the jars have aggressive growth, some don't. Pictured below are jars 1-14, minus #12, who displayed early signs of trych. Visually significant signs and emerged between midnight 9/25 and 5:41pm. Trych is so extreme, it's the bain of my existence. It's mostly what I see with a roughly 30% exception.



Jars 1-4, 6 & 7 are displaying the most prolific growth. All of these are at 5 days. Jars 5, 8-11, 13 & 14 are lagging behind but still have significant colonization around the noc points. I'm not seeing that really good, ropy rhizomorphic mycelium, but it looks healthy. I need to further research genetics and see what i can discover about attributes and phenotypes.

i'm worried some of the jars might have the beginnings of a bacterial presence, especially 5 & 9. There are some grains up against the glass, but I question whether that is just the myc taking its time to colonize the tight spot. No sign of metabolites yet, drains are appropriately dry. Seems to be performing well.

Update 10-7-22

Here are some pictures after casing yesterday 10-6. I can tell it's definitely bacterial, especially from the smell, which is slightly sour/coir smell mixed together. It looks like it's going to colonize and produce fruits though. I just hope the KSSS is fast enough to outgrow any mold that might establish itself too quickly.



Update 2.5.22

After numerous months of playing around I've come to update this entry and keep it current. Trying to decipher my notes is an arduous task to begin with, hardly to mention the amount of work i've been putting in.

So 121 days later I'm comin back to give you the skinny. The whole 9 yards.

Suspected bacterial shoebox decided to fruit. No complaints here.



The spawn performed better than expected given the advantage of a 1:1 ratio. There could have been innumerable factors at play. It fruited in a single cluster at first with the remainder of the primordia nearly fully developed. I know it is common for bacterial tubs to fruit early, which was the case here, and was later lost to Trychoderma post-1.5flush

I do not have a calculated wet and dry weight due to a lapse in my note-taking procedure. Here are some pics of the freshies:




So there you have it. I would definitely call this grow a failure according to my standards. I attributed my baccy spawn mainly to old t3 plates which were kept too long at unstable temp and used haphazardly.
The other half of my problems lied in the fact that my 16qt Presto warped when it was left on dry heat, resulting in insufficient pressure/temp during later sterilization cycles, even though obvious signs were not necessarily obvious.

Thought I'd take the time to finally update this as there were a few familiar faces taking interest. I currently have several projects in motion and will be posting some new grow alongs soon.

Hey guys
So real life happened and I've been away for about 10 days. I'm back to report some not so great news, as well as some decent news.

So these were inoculated 9-20-22 which is what i now suspect to be bacterial agar.
I didn't think anything of it at first, but it makes sense that there's something strange afoot when the mycelium leaves large patches like that uncolonized, right?





So far, 3 of the jars have been lost to trych, which I attribute to a lapse in my sterile technique.
6 of the jars have proven bacterial. 5 seem to be colonizing normally but i still suspect they are bacterial. Here is a shot side by side for comparison.



As long as there's breath in me, I'm not giving up though. I plan to do another run of 14 jars this time with some cleaned up cultures.



I know oats aren't the best and there's a lot of legit reasons as to why, but i bought this whole bag and know it can be done. Going to try millet next.

It’s possible that the tiger drop is at fault, but also
I noticed you accidentally missed venting your PC. Oats already have a rep for being dirty. It’s possible, even likely, that they weren’t sufficiently sterilized. Not venting your PC reduces the internal temp significantly, even if the PSI reads the same.

Either way, let us know what you figure out.

That particular KSSS line went crazy fast. I thought it was white mold at first but nope. Most of the shoes from that cycle just got matted and bloomed on. :nonono:
