Welcome to the Wirelock Challenge!

The rules are simple. You must be the first to go spore -> agar -> spawn -> fruit SUCCESSFULLY AND WITHOUT CONTAM using a ziplock bag as your dish and a length of wire as your only hand tool. The prize, as stated, is a beefy packet of 10 spore prints from user Nichrome, the shape shifting teapot. Second place is a smaller, yet equally delicious cache of 5 spore prints.

Of course things like gloves, SAB, flame sources, etc are necessary and allowed. But the agar must be stored in a ziplock and manipulated ONLY with your self-made wire tool.

Spore prints, syringes, and swabs are all acceptable starting points. Any live or already cleaned cultures are not. All mushroom varieties are acceptable, but it is asked that they grow at a similar rate to standard cubes for good faith participation.


Obviously we are all good boys and girls and wouldn’t lie. This will be done on the honor system. Having said that, treat this like you would any grow log and please take plenty of pictures of your process. Don’t be afraid to over share.

Your first reply to this post will be treated as your grow log. Edit as you go, and I will keep links at the bottom of this page to said post for ease of navigation.

9/18:
Getting ready to pour my bags.. agar recipe used 1000ml H2O, 20g agar, 2g nutritional yeast, 14g LME, and a touch of green for luck 🍀 ☘ I usually use 18g agar, but I thought it would be a good idea to bump it up a little for the bags

I've been procrastinating on this a little bit, but I finally picked a print, RustyWhyte, and found some wire to use.. I'm not exactly sure what kind of wire it is, maybe nichrome.. it's pre-woven, but still rather flimsy and very easy to bend. I went very low tech with it. The only modification I made was twisting one end into a loop.

Too f*in tired and lazy to do an agar session today.. but for those of you waiting, here's a quick picture of one of my bags to hold you over :wink:

Still pretty busy with other projects and running on zero sleep.. so I'll try and transfer tonight, but I'll definitely set some time aside tomorrow to get it done.. here's a picture to keep everyone entertained.



This is actually going a lot smoother than I had expected,.. but we'll  see how the transfers go

Back in the lab, doing my transfers in a minute..

Bags look clean, so I may just take a risk and toss a piece to grain right away :gangsta:

Did a similar roll with the transfer bag, but I was careful to not expose the inside as much


First transfer, cut a small wedge


Second bag, I tried scraping some mycelium with the scalpel end.. definitely more of a challenge to get it in the new bag, but I got a couple workable pieces


Third bag, I tried to scrape with the loop, but it didn't work out, so I just took another wedge


Finally, wedge to qt jar oats/Millet

Taking large wedges was more of a pain in the ass with my wire tool than I had anticipated.. one jar just has a small wedge, one has 2 small wedges, and by the last jar I was finally successful getting a medium sized wedge..

I tried using the loop side of my tool to scrape some mycelium to drop in the jars... it failed miserably :lol:

The transfers I did to grain were so small that things are moving pretty slow. Nothing to see with my T1 jars, but here's my T0.. I shook it a few days ago, so there are a couple similar sized areas of growth on the other sides of the jar

Jar update..

T0 has a couple areas of uncolonized grains.. but on the bright side, I'm pretty sure it's not a sign of bacteria, those areas were a lot less colonized yesterday (toward the bottom and center) hopefully it will fill in more by tomorrow..



T1 jars are chugging along.. I'll probably give them a second shake in a couple days

So, agar in a baggie( i assume we sterilise agar in a media bottle of sorts), germ with a makeshift loop, then what? Grains into the baggie? Or transfer from baggie to a grain jar? Do we just germ in the baggie, then add sterilised grain, then add coir and fruit? .
Are we simply proving that agar can be done in a baggie with a crappy inoc loop? I'm guessing this can be done in a sab? And then transfered to sterilised grain jars, then mono/shoebox to fruit?
Just trying to work out specifics
Also, I'm totally in.

As far as I can tell the only stipulation is that the agar is handled in bags only and manipulated with your self-made tool only. How you grow it out after that is a judgement call for you to make. Unless they say otherwise, in which case I’ll edit it in.

Ugh, I don't have any spores.  I need to order more in a rush! 

I want in if I can get the spores in.

Count me as in!  I have a ton of ideas, and will be starting on the tools and materials in the morning!

Update 9/18/2022

I know we just started talking, but this is the internet so I wanted to show you my tool.



It is made from a peice if 12ga electrical wire I had in the garage. 

I used these items to make it. 
1. Heat glove to protect my hand.  Copper is a great conductor of heat, so a hot spot will quickly get to your hand. 
2. The torch.  I think you could get away with a lot of heating elements.  Copper work hardens so you need to heat it to anneal it to prevent it from getting brittle. 
3. A piece of wood with a metal plate on it to work as an anvil. 
4. A hammer to bang with.  "The hammer is in my pants" (someone please get the reference)
5. File to do final shaping.
6. Nail to use the head as a cutter to cut the wire to length.
7. 400 grit sand paper to make it shine.



I started with hammering it straight.  This is simple. Just lay it on the plate holding it so it can't rotate, and when you go down one side flip it 180, do again, flip 90 do again, flip 180 do again.  This will straighten the wire, and starts the hardening process.  The hammer taps are firm, but not with the intent of flattening the metal.  Thing just starting a nail and your hand is still holding it.  Maybe just firmer than that. 

I placed the zigzag bend in the middle to give my hand something to stabilize the tool while working on either end, and in any orientation. 

This allows the bent end to be a cutter, scoop, flat back (for inoculation), side for scraping, etc.

I heated one end and started hammering a flat section, occasionally hitting the sides to keep it from getting wide.  Reheat to allow it to anneal and prevent over hardening. 

When it is flat and about 1/2" long I bent it 90 degrees about 1/4" in.



I did some final shaping with the file to flatten the outside and make an arrow head shape on the inside. 



On the other side I started hammering a general taper and finished the shape with the file to a point.



I then tested the hardness by flexing the tool slightly to see if it bends easily.  If a section bent I straightened it and hardened it like at the first step.  I hammered the entire thing multiple times to make sure the tool was sufficiently hard.

Then I cleaned it up for photos with 400 grit sandpaper.  Which also gave a light texture to make it easier to grip while using. 

9-19-2022 : filled my ziplock bags.

Nothing fancier than the preasure cooker.

3 different agar recipes in pint jars filled to 100ml to test germination rates on the different mediums.  I made 2 bags for each recipe.  I plan to test a germination strategy based on some research papers I have been reading.  I will detail these later, and report in this the results.  If it looks positive I will do subsequent tests. 
Brown rice flour
Corn meal
Light malt extract



Captains log stardate 22-09-2022 I received spores labeled "The Golden Teacher" on a print from a generous benefactor.  I decided it was prudent to inoculate my bags today to ensure germination had ample time to occur. 

Having fridge troubles in the kitchen region and not wanting to risk a confrontation with the inhabitants of the sector I decided to remove a portion of my experiments for another spore germination attempt at a later date. 

No new pictures.  The bags look like they did before I put spores on them. 

Update 28-09-2022

After 6 days I have noticeable growth. 



One on BRF, and one on LME.  The CM agar is not showing signs of germination despite being a "germination recipe". 

Not a large enough sample size to draw a scientific conclusion, but interesting to note. 

Current status,

2 bags lost to contams. (CM2 and brf1)
2 bags germinated. (BRF2 and LME1)
2 bags waiting (LME2 and CM1)



1 Oct update

transfered BRF2 and LME1 to new bags.  Water agar and LME agar.

I got pictures pre transfer.



I will let the original bags continue to grow, and if all come out clean prep grain in the next few days hopefully. 

17 Oct 2022 update :
I have been able to successfully get clean bags.

3 clean bags.  LME water and a filtered cornmeal.

I also have a SSLC I make with a bit of airiel mycelium from on of the transfers.  It is currently sitting in a QC plate.

I also tried a filtered corn meal.  It is acting like water agar



Oct 30th : overdue update.  After running from initial contamination and learning how to do clean agar in bags.

So here is my update. 

I have 6 grain jars rolling.  4 smalls and 2 larges.  The smalls are a safety net to do g2g onto some oats I just made. 



The smalls are recovering from a first shake after about 15% colonization. 

The large ones are just showing signs of growth.  They were all inoculated by the miracle LC I somehow managed to catch while running from contamination.  I also have the agar I will be putting to grain shortly as well. 


Nov 11th update. 



The grain is still populating. 

The smaller jars might by too dry, they are not populating as fast as the larger jars.

With hope I will go to bulk in the next couple weeks. 

Nov 18th

The 2 large jars are ready to bulk.

The smaller 4 have stalled. 

I will try to g2g them to some fresh oats to see if that wakes them up.



I will post the bulk pics when I get there.

Yeah.. someone asked.

Should be interesting.. I'll be posting some pictures tomorrow after I pour some bags. hopefully I don't pour too hot, I don't think ziplocks will hold up as well as polypropylene :lol:

The ziplocks hold up pretty good pasteurizing casing at 150-160 Fahrenheit. I’ll be posting soon too. Have a good idea for the home made tool and was planning on making some agar tonight.

9/28/22 update

I made up a single baggie (not sure why i only did 1?!) on 8/18/22 (sorry forgot to photograph it) and shot some spore syringe into it.  (APE)

Some contam is in the bag, (trust me, it looks worse than it is..)

Easiest part was to just rip the bag open.. lol


I got my handy wire tool hot and just sliced the agar in half and finagled it into the grain jar. I also melted the baggie a little so theres burnt plastic on the wire tool.. so that should be interesting.


Into the grain jar! Now we wait :cool:

I spawned my grain jar this last weekend! (11/5)

Sent to fruiting conditions today! I wanted to wait a little longer..but i noticed a side pin started. So here we are.

I started noticing very small side pins.. So I took my fruit out of my small dollar store tub and put it in a bigger tub, allowing side pins to grow.
Other than that havent seen any pins..
Guess next time ill use a liner! :P

We got blob pins! I recently moved my tub into my living room because its warmer in there.. I think thats why it was stalling before. Was way to cold!

You can use a loop by squirting some of the spore syringe into a shot glass or other clean dish then use the loop to transfer it from glass to baggie to streak spores onto agar. If syringes are allowed.

It’s usually not recommended to squirt from a syringe to agar directly. Too much fluid. It’s best to take a little drop onto a loop and then streak your plate that way.

Personally I think a syringe should count. If anything it might actually make it slightly more difficult.

I will be attempting this with Ps. Tampanensis assuming that exotics are allowed. (Funny enough, those Tamp spores were given to me by Nichrome.)

I finally got my agar poured.



9/26:
Out of the five bags I poured three contaminated. I streaked the other two with Ps. Tampanensis. No pics because they still look the same as before I streaked them. (I may update with pics of the contaminated bags soon)
