Noob learning and first grow log
#27936960 - 09/06/22 10:08 AM (1 year, 8 months ago) |
|

I am getting materials together to start my first grow while waiting for the spores to arrive.
ToDo: get links to TEKs that inspired my purchase and process decisions.

My first learning experience will be with agar. I will be making dishes to spawn and isolate, and also making slants to start practicing mycelium management and long term retention.
SAB

I am making a SAB from a 105qt bin. I need to go to the store to pick up a larger can to make the arm holes with. I measured my forearms and a tuna can (saw recommended in one of the many SAB Teks) isn't large enough.

This is the agar agar I picked up. It was a much better price than the telephone. Time will tell if it is a mistake to try a different brand.

This is the dried malt extract I picked up.

I will be making it is small batches in this 1L jar. The advertisement stated it was borosilicate, but the jar does not confirm. I bought a pack of 4.
Short Term Goals :
This is not a step by step. Some of these are skills and educational processes to teach parallel skills. Some are for future use skills. Some practices may result in waste.
Agar prep 1 : agar agar + DME(dried light malt extract) Agar+DME Agar Prep 2 : agar agar + DME(dried malt extract) + nutritional yeast
Inoculation of agar with spores (3 for larger Isolation opportunity and to minimize risk of noob mistakes)
Isolation of mycelium. (Dependant on results of spore propogation)
Make "Master" agar based on successful Isolation chains. (Some people may argue that making masters from spore propagation is a waste. I am doing it for habit training purposes, and to buy time to test all successful Isolation for more opportunities at the genetic lottery. I would prefer to do smaller batches of a single genetic chain rather than a large number of batches from separate genetic chains.)
Transfer to slants. (Depends on "Master" counts) (practice for when I start cloning, and for genetic library of pre grow samples. Slants may be destroyed after testing)
Propagation agar plates from selected "Master"
WBS grain prep
Transfer propagation agar to WBS. (6 quart jars) (1 for wbs, 1 for g2g, 2 for bulk, 2 to fail)
Grain 2 Grain transfer. (1qt to 4qt)
WBS to Grain LC. (1 quart jar for practice) Grain lc
Grain LC to WBS. (2qt)
Grain to bulk transfer (4 12qt shoebox:1qt grain 2qt coir (2 from initial grain inoculation, 2 from g2g)
Bulk shoebox grow. (Fingers crossed)
Cloning to agar
Repeat
Sample label and log format: (subject to change)
Goals : 1. Capture full lineage and history of samples. 2. Make "codifiable" sample coding system to make log application (I work in tech and want to make a sample inventory app. I am going to be growing edible and medicine mushrooms as well) 3. Capture image of every sample to visually catalog lineage
Spore (S) unique to strain Isolation (I) Transfer (T) Clone (C) Master (M) unique to S or N to allow truncation when stage is reached. Slave (L) Slant (N) unique to strain
Spore to Master S#-(I#-I#-...)-M# (Isolation is a repeatable value based on how Isolation goes)
Master to transfer format (C#/S#/N#)-M#-L#-T#
Slant format (C#/S#/N#)-M#-N#

You been here reading before you started this hobby huh
I wish I had started out that way. I learn from my failures though. I'm gonna stick around and watch your journey. If you haven't yet found it, this guys stuff just plain works (has for me).
Good luck!

Yeah you're off to a great start, you've clearly done your research. On the SAB, one tip I will give you: make sure you don't cut the armholes too far apart. Mine are a bit wider than shoulder width and it's difficult at times lol.
EDIT: Here is a link to my collection of useful links.

Consider adding some nutritional yeast to your agar, it makes a real difference imo.

I've never used nutritional yeast, what benefits do you think it offers?

It provides nutes for the myc that are not basic sugar. It works well, at least for me. I'll grab a picture of two plates side by side when I have a moment.

I have been reading for a couple weeks lol.
I have probably read 40-50 teks, and watch videos on how labs perform the work.

That is where most of my info came from, with some side influences. Great stuff in there!

I was thinking about that. I saw it in a few agar teks.
I was thinking that less nutrients would cause slower growth. Slower growth might be a good thing as a noob. Give me more time to react to what is happening.
I might try a side by side. I can see a benefit to both methods. Lower nutrients for isolation phases, and more nutrients for inoculation plates.

Here are two side by side pics, both are the same isolate on the same day. The plate with the Nute Yeast is on the right.

Slower is not really better, it will always take days not hours.
As you can see the plate without the nute yeast is slower and looks less healthy, this makes is harder to actually see contam.

Good to know, and great example! Do you have a recipe you like to use?

For 500 mL of agar, I use 10 g agar, 7 g malt extract and 1 gram nutritional yeast. The nutritional yeast contains more complex nutrients that will encourage the healthiest mycelial growth.
TY both for the advice! I ordered some nutritional yeast. It sets my agar making back a day lol. I have some time before the spores arrive. I will probably not use it for slants and masters, but the inoculation dishes will probably benefit from them.
With hope this weekend will be agar making and spore germinating!

Progress update.
I received my spores and lc (po) in the mail today.
I also bought some pink oyster lc for the Mrs.
I made my SAB today. Had to get a larger can to make the holes. My forearms are 13" and the tuna can only has an 11 inch circumference. I am glad I measured first.
I immediately started making agar plates after the SAB was ready.
I made 500ml of agar, and poured 20 dishes. One had a cracked lid, but I assume will still be fine for spore culturing. One I didn't make sure the agar covered 100% of the plate so there is a portion of the plate with out agar.
I will try to use them both for spore sprouting.
I will add pictures tomorrow. I didn't take any while working. I wanted to focus on the work.

I've never measured a gorillas forearm, so not making that claim. Just claiming that a tuna can was too small. Lol

I used a 4in hole saw for my tub and the size was perfect for my arms. Iām not small but not too too overly large. Lol

LOL. I think he's talking about a 13 inch circumference = ~4 inch wide. Pretty reasonably sized forearm if you ask me.

Tuna can is for the johnson, coffee can is for the forearms.

My johnson wouldn't fit in a tuna can, and I'm not sure I would want to try.

But it is only a 2 stroke, so I don't get a lot of work out of it.

Grow log update.
I am currently maintaining 76, cultures.
Of those 2 are controls I use while doing other agar work. They are used to cool tools off after heating, and I rotate them out between jobs. I them store them and watch for contamination.
1 of the controls contaminated after performing a transfer from a contaminated clone of a store bought sample.
12 are PF Tek, updated in my grow log for that.
14 are gourmet. (2 contaminated and being disposed of)
6 are for a grow from a ziplock challenge
37 are PE on Agar going through Isolation cycles.
2 never grew anything and are being disposed of.
3 LC (BRF, Corn Meal, LME)
So I have a lot of material to work with. I plan to put all of the non contaminated samples to grain when the plates are ready. Even the non isolated ones.
I want to learn what makes good mycelium to produce good fruiting, so I will try to fruit it all when it is ready. Fruiting the non isolated also let's me know if isolating is critical, a nice to have, or not important.
This will result in a lot of failure or a lot of fruit to dry and store. Time will tell.
I may have failed to mention a breakdown, the sample count is from counting. The itemized may miss part of the count.

Great start SymPlayTon!! I'm hoping to start my first grow late next after I return from traveling. I like your labeling. I didn't think of an isolation ID, was using Transfer.
Will be watching your progress.
