Did you keep the germination plate and how old is it? Lots of posts about contamination showing up after a period of normal appearing growth. I was wondering if the contamination shows on the agar plates.
Contaminants can’t show up spontaneously, there has to be a vector.

You can also double check your grain by using PF Jars and replacing the upper verm layer with a casing layer upon birth. They'll fruit from BRF. Invitro even lol. Kinda surprised not to see more of that technique.

Cactus soil also can be fractionally pasteurized in a microwave in fraction of the time. I do this with all of my casing mixes, straw, bamboo, yerba, coir.. whatever. Kitchen Cultivation at its finest.

Isn't pasteurizing a rather sensitive technique? I mean one can't check the temps in a microwave. But since it works, do you mind sharing your timings? I want to try casing a few spawn jars with verm and soil and I'm not fond of making all this few-hour-hussle for a handful of soil :sun:

It is a sensitive technique.. however, we don't need to leave anything alive in a casing layer. Not much survives a microwave so pasteurized isn't really a valid term.  I've found the same true for most substrates.

Keep in mind I do everything on small scale. I believe this helps with more than just contamination. For me, it makes everything easier. You just have to do more of it. I have said before most of my failures occur in the late stages. I can honestly blame this only on myself. It's usually because something as too wet. Usually the larger batches suffer the most.

I use gallon ziplock style freezer bags and a small microwave. Everything is brought to capacity and set for 24 hours. Then get as much water out of it as you can. This is the work part lol. Don't be afraid to add a reasonable amount of dry verm to any of it. About 1-2 cups is enough to even out the excess moisture that wants to accumulate. My MW is small.. maybe 1600-1800w. You'll have to play with times on every MW.. even the same model and I have two. The schedule for me is 2 minutes on high. Flip the bag and 2 more minutes on high. Repeat as needed. You want the bag to be a hot potato, but not melted. This might take a little practice lol. Keep clean up materials near by just in case. Usually I repeat the process about three to four times then let them cool. This is gonna sound really weird lol, but I can small when a straw sub is ready. If it really reeks and looks wet and compact.. it's not done or it's still too wet. More verm and cooking is needed. The straw should be moist, but not soaked. It should also be somewhat fluffy and not clumped.
I have two bags at home waiting for spawning. I'll try and remember to post pics.

I think I'll adopt this approach to try narrowing in on my semp contamination.  I like the idea of using the jar as a way to cut out the root cause of contam 50/50:  If you see the contents of the jar contaminating while fruiting it, you know that the source of contam is not related to spawning to a tub, etc. but rather something with the culture isolation or grain prep.

In my case, I believe the root problem for me will come out to me culture isolation but I'm going to use this strategy and find out.

Example from this week-end to illustrate (so I don't keep posting the same photos).

Photos from above, nice fruits:

And here's the shitshow underneath:

This jar was inoculated with a seemingly clean LC. I mean, I've tested it on agar, and it looked perfect! But the result here says otherwise. Or I have a filter problem - I've just changed 18 of them and will see soon. Or I have a sterilization problem, I bought tape changing color when it reaches a certain temperature, PC'ed some jars this morning and will see the result tonight.

If none of these test come back positive, then I'll start to think some contamination is free-riding the mycelium and not showing up until put to grain. Maybe a crazy hypothesis.

Anyway, I'm still debugging... patiently.

Bummer, made 2 LC's .. one of them turned out to be contaminated, didn't check for cleanness on agar before I inoculated, should have done that first. Didn't observe any cloudiness.. both broths looked clean, behaved the same.. so I "assumed" they were fine. Learned my lesson here.. lost 3 small bags to trich.
:fffuck:
The other 3 appear clean, jar also didn't trich out, so hope my LC technique was good and the broth just bad from the start.

Yeah mold in lc is near impossible to spot. At least bacteria cloud the whole thing up for you.

Yeah, I added 3 wedges from 3 different plates to each LC, ofcourse a vector for contam but with sterile technique it should work fine. didn't stir the solution the first 3 days as I noticed myc just emerging from the wedge. I saved the agar plates, which looks good. So I think it was just bad luck, will redo the process. If I fail again, I'll move to the josex'poke.

I'm not going to say normal lol, but it does happen once in a while. There used to be some debate as to whether or not it's a sclerocia formation. Not sure where that ever ended up.

Here you go. Microwave Straw.
Notice how the bag is like a pillow. Even cooled.

Nice and fluffy. Comes apart with a shake of the bag. This one went through six cycles of two minutes each. Nice mild odor now. No pooling water. Ret to go. :uptosomething:

So here is my trip report from yesterday. Went to the beach to go fishing and took a small dose of 0.7g. I ate a sandwich before this that I think dampened the potency so I'll do 1g on an empty stomach next time since some friends I gave them to had a wonderful time on them with that amount a few weeks ago. Anyways I consume them and make my way to a pier and start casting. About an hour goes by before I start feeling anything, then slowly the effects start and the water looks way more vivid than it did earlier. The patterns of the waves and the light glimmering off of it were quite mesmerizing. But that was it as far as visuals went. After another 2 hours of chilling on the pier (and not catching any fish,) my friend and I went to this ice cream shop close by. We went back in the car to eat it and the taste and feeling of the sorbet I got was very pleasant and blissful especially when complimented with the music we were listening to. Throughout the trip my tactile sense was increased and held onto this soft pillow I had, this happens whenever I trip on anything though. Made our way back home and cleaned off. Overall I liked that I didn't get this fuzzy faint white noise feeling I get in my brain that I usually get when I take cubes or acid. I could for sure step up the dosage a bit and go from there as it was a vey mild experience, I just didn't want to accidentally take too much as I was out in public.

I can`t recomand it enought. It`s a total game changer. You can inject  through the SHIP, no need to open the LC bottle.

Sounds very similar to Tampanensis on 0.5g to 1g. Thx for sharing the trip report.

I have used it before, but prefer agar. If it contaminates, it means I've messed up in my sterile technique somewhere. I rather redo it again, be as carefull as I can be. If agar is always good when working with attention, then so should a LC.

Getting a little frustrated at no fruiting. This is try 4. Looking good yeah? Looks like knotting to me. I tried using a bag this time to let it colonize, then opened it up to fruiting conditions, been 2 weeks now. Previous attempts were shoebox, I’m theorizing they dried out so tried a bag.

Been keeping it simple with straight coir and same conditions as my cubes. No casing.

I haven’t given up hope on this one but I’m starting to wonder if it’s the genetics I received.

Previous attempts everything looks great and healthy but nothing ever happens. ¯\_(ツ)_/¯
