This podcast is awesome and anyone interested in psilocybin chemistry should listen to it. Dirk's English isn't perfect, but the information is the newest available, directly from one of the world's top psilocybin researchers.
One of the most interesting things I learned is that psilocybin mushrooms don't have much psilocin when fresh - they contain enzymes that quickly phosphorylate psilocin back into psilocybin. When damaged the psilocybin dephosphorylates into psilocin and is rapidly polymerized into blue polymers. Dirk thinks that the point of psilocybin is a stable way to store the psilocin monomer, and the point of the whole chemical pathway is that the blue polymer is a defense mechanism that inhibits slugs or insects. It is interesting that the defense mechanism itself is inactive in humans, and it's the intermediate molecules that have the psychoactive effect. None of this is proven but it makes more sense to me than any other explanation I have seen so far.

I liked how he related psilocybin to a hand grenade that's stable just so long as the pin isn't pulled.
What an interesting chemical!
It will be interesting to see what applications humans will use psilocybin for in the future.

Glad this info is being spread more. Felix and his team from Miraculix labs out of Germany first propagated these results I believe(don't quote me)
The biosynthetic route is well established now, not only that Psilocin is not produced independently of Psilocybin but rather is the result of a degradation byproduct(esp if seen in test results) the idea is the more psilocin that is seen in test results generally occurs when there is a greater level of psilocybin with potential to break down during the dehydration process.
Test results that show psilocin in samples is the result of poor testing methodology or a degrading fruit body(for psilocybe cubensis)
Also as you mentioned that blueing is not always relative to potency, but rather a slew of other factors including pigmentation, density, damage, cell wall structure and enzyme content! What's really interesting is psilocin doesn't seem to be as unstable as thought if preserved within the fruit body, in terms of Pan Cyans, recent test results are showing elevated levels of psilocin, some nearly matching or higher than psilocybin content, this leads us to believe that it is remaining at least somewhat stable within the fruit body, and being produced independently.
The current data is only for Psilocybe Cubensis. In terms of Panaleous Cyan in recent test results show that likely has a different biosynthensis route and enzymes. As with a cubensis you should never see elevated levels of psilocin. Also, recent research hints at other species of psilocybe potentially producing their own unique novel compounds.
"Additionally, in Psilocybe cyanescens there is a second monooxygenase (which we name here PsiH2) as well as a third MFS-type transporter, all of which are expressed at the RNA level (Fig 1a). These findings hint at the exciting possibility that mushrooms from the Psilocybe genus, and particularly P. cyanescens could be producing novel psilocybin-like molecules."
