For what it's worth I use dedicated plates for cooling my tools when I'm doing agar work so my plates look a little nicer. There's no reason to throw out plates that aren't contaminated - even if they're red

I've never experienced any issues with a contaminated cooling plate that I'm aware of, which is actually a little surprising considering I keep my cooling plate open in my SAB the entire time I'm doing my sterile work. I do usually wrap my cooling plates at the end of a session and keep it around for a couple weeks just to see if any nasties pop up, especially if I'm sending any agar to grain/LC. I'll label it with the date and what I was working on in that session just so i can keep track of things, similar to a lot number for quality control

I've always cooled blades in the edge of the reciever dish I'm transferring to. I don't think I've ever seen that cause contamination either. I'm curious how much of a contam vector a blade can be really. In front of a flowhood unless you touch the blade to a dish you know is contaminated or bump it against anything it probably stays sterile. I've messed around doing multiple transfers from the same dish without re-flaming the blade each time and haven't seen it cause a problem.

When I do my transfers I use a Stainless Steel drinking straw to punch perfect circles. I'll flame sterilize the straw once, cool it, punch-cut all the transfers i want to take, switch to my xacto (which is also only sterilized once per plate) and make all my transfers. No issues yet as far as only sterilizing each tool once per plate.
I should also point out that I wrap my tools in foil and sterilize them in the PC. I won't run the PC just to sterilize tools but any time I cook agar/LC/grains I'll toss the tools in. Can't be too clean

I tried the stainless drinking straw thing but i noticed when trying to delicately isolate an area where rhizo connections are, it's lack of sharpness tore the connections. I tried sharpening it with my bench grinder but that made the cuts then stick inside the straw.
Lately I've been experimenting with these plastic handle #11 steel blade disposable scalpels. I made little metal caps for the blades, wrap them in foil, and pc them laying flat on an aluminum disk so they dont warp. I don't flame them and they stay perfectly sharp. I've also found a company who claims their plastic handle ones are autoclavable. If they don't warp they could just go in a jar individually wrapped and not need to lay flat.

Update on my four transfers! Plus two more since then... Lol




The little guys have a lot of condensation but they are all growing super fast!!! My original dishes are almost grown out too.

They look healthy. Damn red dye is getting it's funk on. I can see the growth is getting odd. I'd definitely use a different color in my next plates before it gets any stranger.

Haha yeah I threw the red coloring out, just burned my last red plates on those last two little guys. Next batch is going to be blue lol
