Well I 100% agree what I suggested does no good to clean contam, I was just curious about the myc behavior.
Interested in more experiments and results from you

Hey Rotn, so did you ever get an opportunity to try this with a sample of contammed myc onto a clean WA plate with a clean nute sample nearby? Or with adding the nute sample a day or two in advance of the myc transfer to see if it aids in directional growth?
This thread has been a great read for weeks, and Iโm totally putting it to use this weekend cleaning up a bacterial Rustywhyte plate that I cannot get clean after multiple transfers. Gonna go back to the T1 plate that is developing primordia even though I know it to be bacterial.
Plus, after being out of practice for a couple years, I took a dozen clones today from a small MS grow and highly anticipate the need for some serious clean up. Method felt a little rusty. Gonna pour some WA plates tonight to have on hand for the transfers.

I haven't yet, but I plan to start some new plates with different versions of the experiment sometime in the next few days..
I'm a perfect example of that "pokemon" thread someone posted recently.. I've got over 10 cubensis varieties, 5 non cube active species, and 2 gourmet species all running at once (not to mention the different clones I'm testing of each one) ๐
I was keeping up with it really well at first, but I'm starting to get a backlog of jars that need spawned..
I plan on spending the weekend spawning jars, and hopefully I'll be back on track and getting back to some agar work/the WA project sometime next week. I've got some other cool applications I'm testing out and plan to post the results soon, so hopefully I can keep the thread interesting 
I'm glad to see more people giving WA a try can't wait to see your results

The nutrient experiment plate has grown out.. I didn't notice any change in growth once the mycelium reached the sample..
I accidentally left the plate out of my stacks, and it developed a bit of condensation. I'll post a picture as soon as the plate clears back up.
Also, I'm cloning some wild fruits later today.. I plan on sending tissue samples to both nutrient and WA plates, I'll do a side by side comparison of how long it takes me to achieve clean cultures.

I did the same thing once.. I went in to auto pilot when prepping my plates and forgot, until after I finished pouring, that I was supposed to be doing some WA plates in the batch.. I cant wait to see how things go for you once you've got the right plates 
I'm also waiting to test it out.. after ID'ing my finds, I decided to wait. I'm going back out tomorrow and over the weekend. Even if I don't find anything interesting, I'll still pick something to clone just for the sake of experimentation..

My first water agar plates (not pictured) have been sitting for a couple days now. So tonight Im going to take the tiniest transfers I can cut, try to transfer the primordia off the below RustyWhyte T1โs that Iโve been ignoring for a bit. Going all the way to T4, I have repeatedly been unable to get the myc away from bacteria when using PDA.


Itโs a variety Iโve wanted to grow for a long time, but havenโt been able to get anything clean enough to put to grain. So Iโm pretty excited to try this.

It actually did end up showing different growth that I didn't expect... there is a ring going around half of the plate right where the nutrient sample is, where the mycelium is a but more pronounced and is has aerial strands

Cool, like itโs demonstrating the way that the myc as an organism acts as a single network of redistribution. Once it reached the nute sample at 12 oโclock, all the myc at that same radial distance from the center all started getting boosted nutes at the same time and produced aerial together all the way from like 10 or 11 down to 5 oโclock .

Is there more aggressive growth from 10 to 2 around the nutrient area? Iโm seeing the growth from 9 to 12. Iโm surprised the growth isnโt circular if the nutrients were leaching into the water agar.

Nearly impossible to photograph, but I am getting visible growth from my RustyWhyte transfers onto the surface of my water agar plates!


I took the tiniest samples of primordia (in hopes of getting a culture suitable for fruiting from the original MS T1) with as little of the bacterial agar as possible from the donor plates, so I wasnโt sure how much nutrition the samples would have to grow out on the new surface. But it looks like theyโre doing okay.

Same 3 RustyWhyte plates. Looking to my eyes like a success.


All previous transfer attempts had ended up snotty and bacterial no matter what I did. But these look clean and uniform, from what I can see.
True test of course will be transferring back to nute plates. Will put some outer edge growth to GWA this weekend.
Thanks again for the thoughtful write-up, Rotn. Iโve been so pleased with this result, I took tissue samples from a bacterial MS bottle-grow of Ghost yesterday, and cloned straight to WA; and am working a couple FP+ clones that looked just a little iffy. A good method to have added to my toolbox.

Hell yeah I've got a good feeling this is going to do the trick and get that culture clean for you.
My one recommendation is to keep the WA donor plates around.. there have been 1 or 2 situations where the culture was "cleaner" but I transferred back to a nutrient plate too soon.. since I held on to the WA plates, it was an easy fix.. I just waited a little longer and took took another transfer once the WA plate was almost done growing out.. those plates are looking good, I think you'll get your desired results on the first attempt, but it never hurts to play things safe and have a backup ready to go.
Can't wait to see how the culture looks when it's back on a nutrient rich plate ๐ค
