No sign of Rottn? I've had several varieties of cubes and woodlovers on water agar since Last fall. Unfortunately, I don't know how to read the mycelium. Some have white rings, some have cloudy halos, some have brown rings. I'm never confident taking a transfer because I don't know what I'm looking at. At what point are you sure the mycellium has out run the bacteria?
Taking pictures of water agar is a challenge in itself. Unless, a person is set up with back lighting. Does anyone know of another thread that shows mycelium on water agar?

I'm not sure. But the theory here is that the bacteria doesn't grow out from the transferred piece. So if you grab some myce that has grown out towards the edge your going to get something clean. I suggest transferring while back lighting in a dark room under magnification if possible.

This thread is an amazing resource. Noccโd up a few deli cups with a TYC print labeled โdirtyโ the other dayโฆ I could see bacteria on day two haha, over half of them. Nice to have this in my pocket. Thanks to all you guys!

I just got some cultures and spore syringes. Can I use water agar or should I add a tiny bit of karo? I have been making potato flake agar and it works but I inevitably get slime.

Usually water agar is a tool used when you have a bacterial infected culture. While you can probably germinate spores on it. I would personally germinate on regular agar first.

Just remember this tek wont help with mold contamination cuz it's mycelium and will grow on WA. This tool is only for bacteria.

It's probably gonna look real wispy and difficult if not impossible to distinguish from the mycelium you want. If you think a culture has mold, regular agar would be better at feeding it so it shows stronger characteristics that allow you to transfer away from it easier. Just my opinion.

Well, I have a few plates that develop wispy mycilium around 60+% colonization. One of these days I'll figure out how to take pictures of WA from my phone. Trying to get back lighting and a background that doesn't show light fixtures/wallpaper/windows etc...has proved frustrating.
