I'm still a youngster (early 30s)... but, yeah.. I really started enjoying agar work once I got my FFU.. it always felt like more of a chore in a SAB

I was joking, in my book WA is the new standard for cloning. It can not only cut down the time to get something good but also reduces the number of plates needed. I am all in when it comes to saving resources and simplifying the workflow.
It's an awesome tool.

It IS an awesome tool.. if I didn't think so, I'd never have done the write up.. but I still feel strongly about the whole time and place thing...
Sometimes it's better to use a bazooka to take out a big ol' contamination monster.. but pulling out the bazooka to blow away any tiny amount of contamination just feels lazy..
but everyone has their own cult style and overall goals.. some just want fruits, some want to enjoy the whole process... however someone chooses to use the tool is completely up to them... as long as they find the tool useful, I'm really glad that I did this write-up and brought a little more attention to it

Yeah I will only use it for cloning and the rest will be regular plates for germination and cleaning germinated cultures.
On another topic, would you think that water agar could be used to get to a mono culture faster since the growth is so much thinner when is spreads out? I think that might be another application for this

Don't have time and space to get that fancy atm......but one day maybe. You can use a microscope to confirm a monokaryotic culture but I doubt that you could confirm a mono-culture as in a culture made up by one single strain.

Just cause it's a culture that doesn't sector and has even growth all around does not mean its a mono culture.

I'm on a mission to start using up some of my ridiculously large plate collection.. So, I decided to do a side-by-side, plate to grain, comparison using 2 fully grown out Phobos plates.. One plate is water agar, the other is MYA. I decided to do 3 jars from each plate.
Quick picture before getting to work:
Here are the plates I'll be using
Plate 1 (MYA- cut into 3 wedges, leaving the center and outer ring behind:
Used plate 1 to inoculate 3 jars of oats:
Here is plate 2 (WA- cut in a similar pattern to plate 1)

Used plate 2 to inoculate the other 3 oat jars:

I plan to post updates comparing any differences in the length of time it takes for the mycelium to jump off, how long it takes the jars to colonize, etc...
I may continue to update through the first flush, but I'm not really sure if it's relevant to this thread or not continuing to watch the grow until completion, or if it should stay limited to agar work and jar colonization

im excited to see the results rotn!! i feel like having the jars get to 100% colonization would be enough but if you feel like posting some first flush comparison pics i wonโt complain. youโd think once the grains are colonized it would be about the same tho

I think it would be cool to see the conclusion go all the way to fruiting, if the grain colonization doesn't give a significant difference first. Either way I think it pertains to WA. This is a great thread!

So, it's still early in the first test run, but it looks like the WA plates may end up taking longer than the nutrient plates to jump to grain (or this batch may not jump off at all)
Note that this batch of WA plates were around 3 months old, so it may be an indication that the mycelium can't survive that long without food (makes sense, right )
I'm going to keep observing to see if anything happens.. I also plan on starting a new batch of WA plates soon, and re-running the test with fresh plates.
So far, my hypothesis is that mycelium on WA plates doesn't stay viable very long.. so once your plates grow out, be sure and take some transfers before waiting too long.
This also gives me an idea for another WA test... I plan to make a few extra plates in my next batch. I'm going to put them in cold storage, and take transfers once every month or so, to see how long the mycelium stays viable in the fridge.

That's interesting stuff. Nutrient agar transferred to a water slant grows out some then stops when it runs outta food but stays viable for a yr or two
