I second that. For a while I'm using agar with 1% LME for everything including germination with not problems. Thinking about lowering it to 0.5% and see how that performs.

This makes sense in principal. I assume that the mycelium will direct growth towards the nutrient source due to nutrients leaching out from the MYA agar transfer. Makes me think that this will work best if you make a relatively soft water agar and up the nutrient strength in the piece of agar that is suppose to attract the mycelium. Maybe also experiment with preparing plates in advance and let them sit for a while to give the nutrients more time to leach. You could also try a dollop of BRF past on one side to lure the mycelium.
I feel like slime molds are more vigorous, mobile and directional when they are surveying their surrounding but there is no reason why other mycelium can't be directed when a gradient in nutrient concentration is present. Are there any published papers on the topic that do include a description of the experimental procedure with recipes?
Thanks for doing this. Will be interesting to see what happens.

Was going to use water agar for clones as well and accidentally prep all my no pours on the last run with LME. Bloody hell

OK, it's happening, 6 out of 25 no pours currently sizzling on the gas burner are water plates. Will have to wait for some of the new pan varieties to clone and they are not even on grain yet. I might take a look in the garden and see if I can find some random LBM to clone instead.

Ok, I forgot that my little Pan Wild Coast straw bottle started pinning so I took one of those to WA. I'm already in the process of cloning this variety but only one of the 4 transfers I made is showing some growth and as usual it does take a while before the mycelium gets out of the goop zone.
This is the transfer on normal agar made 6 days ago.

and this is the pin I placed onto water agar just now, straight to plate with no prior cleaning/washing.

I'm sure the first sample will work out eventually but I'm really curious to see how strong bacteria is going to flare up on WA.

The pan pin is showing good recovery in the mycelium department while bacterial goop doesn't seem to thrive.

That is already a win in my book. Working better than tee agar so far.

Honestly using water agar is like cheating, where is the craft and art in this? No need for taking great care to cut the tiniest sample over and over again just to find out there is still a contam riding along. I was throwing this pin onto a water agar plate and taking transfers when it had barely grown out 10mm just to get 4 pristine clean plates on the first try and I could probably have gone straight to LC with this making this whole thing soooooooo easy boooooooring. You young folks are missing out on a painful but character building experience.

What challenge has life got ready next? Painting by numbers maybe?
This is utterly ridiculous.

I was joking, in my book WA is the new standard for cloning. It can not only cut down the time to get something good but also reduces the number of plates needed. I am all in when it comes to saving resources and simplifying the workflow.
It's an awesome tool.

Yeah I will only use it for cloning and the rest will be regular plates for germination and cleaning germinated cultures.
On another topic, would you think that water agar could be used to get to a mono culture faster since the growth is so much thinner when is spreads out? I think that might be another application for this

Very interesting idea.. unfortunately I'll have to do a bunch of research before I'm able to test it out *challenge accepted*
If you or anyone else wants go get a head start, while I'm slowly trailing behind on this one, I'd be really interested in seeing how it goes
Do you know if you need a microscope to confirm a monoculture? (it sounds like one would be necessary)
Don't have time and space to get that fancy atm......but one day maybe. You can use a microscope to confirm a monokaryotic culture but I doubt that you could confirm a mono-culture as in a culture made up by one single strain.

Cloned to water agar. Took tissue from inside the caps and also dropped almost a whole cap as well. Cut off and placed with non-sterile scissors. Let's see how forgiving WA is.
