What makes you think they are monokaryons? Do you have some way of dealing with single spore or just serial dilution?
Not doubting you or anything I just know very little about this aspect.

Yeah, I had a few pop up on the 2nd flush of an APE cake... but they didn't make it to maturity for some reason.. well, they did, but they didn't look right.. maybe I let them go too long 🤔 😕

Nice prints. Recently, I have been setting my caps on the wire rack and printing on foil beneath them. It seems like if I can keep the gill material from touching the foil, the print is cleaner. Especially if I also employ the “second print” technique.

Thanks 😊 I'm taking second prints right now. I let both first and 2nd go for 24 hours. And if the caps are small I double or triple up. If anybody wants these, I'll probably do a giveaway tomorrow after the 2nd prints are done.
I was reading a thread about the wire rack method you use the other day and was thinking about trying to use one of those mesh stands that are used with bunsen burners and printing directly on a plate with that method
I know it would be a TON of spores, but if you did a first print on foil and then only let the one on agar go for an hour or two thoughts?

Next time you should check out the water tub fruiting chamber. In my experience, it is much more effective at holding the correct humidity for cubensis cakes, especially if your SGFC is not packed full of cakes. This will eliminate or greatly reduce the amount of misting needed, which makes things much easier.
Good job though, those are some pretty mushrooms, and welcome to the Shroomery!
@Rotnpins I have never tried printing onto agar, and I feel like it would make the resulting germ plate almost unmanageable, but it is cool. When I do first prints, I do keep them for myself. I kind of like cleaning up dirty prints on agar. It is good practice for working with field prints from outdoor finds. I reserve my “cleaner” prints for giveaways and such, as I often never know the recipients intent or agar skill level, and always want to send the best I can. Please do a giveaway. Giveaways are one of the most wholesome parts of this community, and I feel like the mushrooms want us to spread them far and wide. It is the Lord’s work. Keep up the good work Rotn!

That big one gave me the biggest cap I've had thus far. Printing now. That's a wide mouth half pint for size ref.

I think Fahtster does something similar using a jar, a wire holder, and all inside of an oven bag. I can’t remember his exact process, but he is well know for having the cleanest prints and swabs around, despite his unconventional techniques. In my limited experience working with Faht’s samples, they may be the only spores I would ever consider taking straight to grain. I’m paranoid, so I wouldn’t, but fern plates from his giveaways have produced the cleanest germ plates I have ever made.

Distilled water is NOT sterile. If I were to experiment with that method I would use distilled water but it must still be sterilized. AFAIK when people make spore syringes they'll fill a syringe with distilled water, wrap it in foil, sterilize the whole thing, then shoot the sterile water onto the print, scrape the spores with the needle then suck the solution back up
