It can't hurt to watch the plate to see if you get other germinations. But I don't see anything that could be cleaned up out of that spot.

The little white dots at 7 o'clock right beneath the yellow should be mycelium.
That's where i scraped the scalpel. You can see the cuts in the agar if you look closely.

Could you take a better pic of those spots? I can't discern them easily, and I am curious. If this orange spot is away from the spot you inoculated, then you can definitely save the plate, as bacteria don't spread very quickly like mold. I thought that this spot was your inoculation point.

Edit. Right 7 o'clock. Yeah looks clean, I usually let things get a little bigger before transfer but I can see why you wanted to get it out of there as soon as possible.

It’s been a minute since I’ve grown anything on grain, and maybe this is good to go, but the way it’s looking seems off to me, maybe?
This is p. natalensis. Liquid culture, tested twice on agar, looked good on agar. PCd grain jars for two hours, inoculated grain jars with 10ml of LC a week or so ago. They were showing nothing a day or two ago but they’re all showing now.
The “hairiness” of this seems off to me but maybe it’s nothing. If it’s of any relevance they’re in a 78 degree room.

Does mycelium growing on agar benefit from a light source or is it just the same if you keep it in a closet?

Yahra did you streak it or just put a drop in the center?

I flamed my scalpel until red hot, cooled it down in the agar and scraped a few spores from the print onto the blade.
Then i rubbed each side of the blade against the agar.
Pretty much in the center. No fancy streaking in patterns involved.

Ok, now I see the spores. It doesn't look good, but I would wait, hoping to get at least some myc showing, before the bacteria takes over, and transfer from the best looking edge using a needle point. Frankly, I believe that you will manage to get a transfer out of this plate. Key word, "believe".

I totally get that, first plate is special.
However, some of the best advice I can give about cult is be ruthless, absolutely cutthroat. If a culture doesn't perform perfectly, axe that motherfucker. jar look sus? Chuck it. Tub taking way too long? Dump it and start over.
All that iffy stuff just takes up space, resources, and time that could be going to performing genetics and clean cultures.

That's a toss, right?

Since no one answered, i rephrase my question.
Does it make sense to wait for mycelium showing up and try a transfer or is it futile since contam has a massive head start?
That looks like very slow moving/non motile bacteria (likely streptococcus from the look of it) and if those spores germinate the mycelium should be plenty easy to transfer cleanly in one to two moves if you do well.
I deal with nastier shit all the time when working with wild tissue/spores.
Just don't let any condensation liquid roll across that bac colony. Any liquid will transport them and help them proliferate.

Gamechanger for me taking pics of plates was to put it in manual mode and move your camera closer or further depending on manual zoom you have set. The idea is to move the camera so that it focuses on the agar surface not the plate. With auto focus it always focused on top of plate for me.

Trying to work with what you have is how you get the experience that makes you wisely ruthless like Mr Piggy. I would keep that plate and see what I could do with it -- while also accepting it may well be a dead end.
It works out or not, either way you learn something, and if it craps out you haven't lost much.
