First time swabbing a mushroom. I swabbed the gills of a big PE fruit whose cap was nice and open. The swab turned dark which I assumed meant spores were now all over it. I let it dry then bagged it up. About a month later now, I used it to streak a plate. I never got any growth aside from a single dot of contam. What happened?

Yo Aspect!
I used to try streaking with swabs and never ever got growth regardless of how hard/soft my agar was, then I starting either cutting a small piece off or now using flame sterilised tweezers left to cool a little just twist or pull a small piece off and stick in the agar, worth a shot.

Patch off odd looking myc, I've never seen cobweb mold in real life, but this looks like..... a cobweb... it's noticeably grey in comparison to the regular looking cube myc. I've
isolated the jar (I didn't plan on growing in tiny agar jars, it just happened) from the rest of fruiting chamber for now, what do you guys think?

I refer to this almost every day.
That looks like Rhiz or Muc.

Same thing I am growing in one of my jars.

I know it's late in the game to be asking this, but here goes. All of my grain jars have a 1/4 inch hole in the lid covered my microppose filters. Do I still need to keep my lid loose while pc'ing?

I just put water up to the bottom of the jars, or just a little higher, to be safe. Even when PC'ing for 3 hours, I've never run out of water.

Thank you! I'm glad to hear it! I'm a little less worried now

No, you can tighten it fully. The hole will distribute pressure in the jar as it's built up.

Hi there

The only whole grains I could get around here were wheat. Could anyone point me to a wheat prep tek? Or can I just use the No Cook WBS tek (i.e. soak overnight and then let dry in colander)?

I'm having some contamination issues that I'm trying to figure out. I switched from an instapot to a proper PC that can reach 15psi.
Grain prep: bring water to a boil, add wheat in brewer bag/paint strainer bag. Once boiling, set a timer for 20minutes. Cool on rack with fan for 1-2 hours or until outside is dry to touch.

PC: Load bags- I am loading about 3-4L of material into a bag, folding with filter protected, and pressure cooking (after 15minute vent) for 3-4hours at 15-17psi. My bags are elevated out of the water. I let the bags cool overnight in the PC.

Next day: I spray bleach and wipe down my vertical flow hood. 20minutes later, I spray alcohol and turn on the fan. I load spray down agar plates, my scalpel, and bags and load them into the hood. The bags seal them selves because of how they are folded.  I cut off the fan and cut the agar into a few square and drop into the bags. I allow air to enter into the bag, fold over the top and wrap a rubber band around it tightly.

I noticed one of my bags today still had trich and looked wet. Am I loading too much into the bags? Under hydrating my grain? Not every bag is having this problem that I can see so far. Load less into each bag? Thanks in advance. Here are some pics to help visualize what's going on:

No visible trich, but grains look bacterial from guide. This is P. Natalensis inoculated 10 days ago.

Visible trich starting (or bruising?). Spawn not looking great. Machine elf inoculated 7 days ago from agar.

P. Semp spawn bag inoculated 3 days ago with agar wedge. Grains look good here.

P. Nat plate pin agar T3. No visible trich. Grains look good.

Any advice would be greatly appreciated.

If Its brain waves are interrupting your normal daily thoughts. If going about your normal daily tasks and you hear ‘harvest us! Harvest us!’ Then it’s probably time to harvest.

In all seriousness it’s normally when everything stops growing visibly for at least a few days.

I found some old dried up cultures which laid around for over a year at room temp. Is there any way to recover these or are they dead?

Regular golden teachers. From my own spore prints so I'm sure. They look like PE blobs.

Best way to find out is to take some transfers to new plates.

I was unaware of the insta pot issues. Are you sure it's not your flow hood?

The first bag looks a little wet but I can't say for sure that it's bacterial.

The second bag looks a little off for sure. I see the trichoderma on the bottom like your talking about. That one is history...

That seems like a long sterilization time as well grains should be plenty sterilized. I mean 2 hours in an insta pot has always been sufficient for rye...

I say this because flow hoods can give you a false sense of security and can make you very frustrated if they don't work correctly.

Since you open the bag and let air in I'm curious if your also getting some other spores in there at the same time?

Do you know for sure it's not a flow hood issue?

Are you sure you aren't being too critical of your work? I don't see anything major that sticks out to me other than bag #2 has some trichoderma.

I'm still learning myself though.

What is this at the bottom of my jar? I got this weird yellow black thing. I have read around and got mixed feelings, worried about possible contam.
First BRF jar, 200ml.

2 days after fully colonized, looks almost like a face lol.

5 days later (looks like small pins?)
