Slow colonization on Rye
    #27810826 - 06/08/22 01:04 PM (2 years, 7 months ago)

This is only my second attempt at growing.

I have 13 jars that I attempted to inoculate via MS syringe. I soaked the rye grain for 12 hours with 1 tbsp of gypsum. Boiled for 20 minutes until plump but not bursting. Steam dried until they were dry to the touch. Sterilized for 100 minutes @ 15 psi. Used a SAB and isopropyl to sterilize myself and equipment. I keep my room at around 76°F.

My question is, it's been one week now since I've inoculated, without even the slightest sign of mycelium growth. Am I being impatient, or should I be seeing some amount of growth by now?

It sometimes takes 2-3 weeks for spores to germinate.

Be patient.

It is best practice to send spores to the updated pf tek because most spore syringes carry some level of contam.  When you shake the rye you will inadvertently spread bacteria all over the jar.  It will probably still fruit just not as well.

Or you could learn agar and clean up the syringe in agar then drop the clean culture to rye that works really really well for me!

My first attempt with MS syringe went miraculous. 8 jars, and none had any contamination.
I attempted to make some agar plates a few weeks ago, and we're completely taken by contams. So I figured I would try the method that gave my great results the first time.

They will most likely be ok. Probably just need a little more time.

Yeah you gotta do whatever works for you.

Good luck!
