Once agar plate transfer is cleaned?

Alright so my question is, after successfully transferring agar to the point where you have a clean plate with no contams visible. About how many jars/bags can you inoculate with that clean mycelium. I’m talking about your standard Petri-dish

It only takes a tiny wedge to inoculate spawn, so depends how many transfer sized pieces you can make from clean sections of the plate.  More clean inoculant = faster colonization.  If you're going to do bags, I would suggest using a grain master or LC to keep colonization times down to 10-14 days.

Some people use 1/8 or 1/6 of a plate for one half pint or 1qt jar but I use either half pint or 1/3qt jars so they're not that big and I use wedges around 1cm x 1cm (.5" x .5") in size. I also drop two or three of them, depending on how much of it I have left, in there to speed things up.

I used to do wedges around 1/6th of the plate per jar. Got bored of the colonisation times and started using half a plate.

If both your agar and technique are good you won't have a problem with half a plate. Heck, a whole plate if you really wanted.

I like to go with half a plate so I can get 2 jars out of each plate that colonise quickly. I always have a lot of plates around so it's not an issue for me. Faster is better for me whereas more spawn per plate is better for some.

I slice a plate into 4 and each wedge goes into a different jar

I usually do one final tiny transfer then drop the whole plate, then shake after 4-5 days.

No point in dropping a tiny wedge, torturing yourself waiting for a jar to colonize, unless your into that kinda thing :badslave:

I will take an entire plate and slice it into several sections like a pizza then drop that into a jar that is only maybe an 1/8th full of rye.


Once I see the the mycelium take hold and jump off the agar onto the grain I shake it up. It will colonize fast and you can then g2g transfer jars with that.
