First Agar Grow - Identifying Mycelium?

Hi there,

Hoping you guys can help.

When I did my last Mazatapec grow, I took a spore print as I wanted to try growing from agar the next time in order to get a cleaner grow and better yield.

I have put spores in 2 petri dishes, photographed.

The one on the left appeared to be myceliating after a couple of days, but now I see the dark green growing in the middle of that white patch, which I imagine is contamination. There is also a pink blob, which is clearly contamination.

It raises the question: how do we distinguish between the desired mycelium (in this case Matatapec) and contamination?

The one on the right looks cleaner, and has some white patches growing. How to tell if they are what I want?

Finally, the one on the left... do I throw it away or see if any of the other white patches that are softly emerging are any good, in order to transplant to a new agar dish...?

Let´s call it top and bottom plate for less confusion :wink:

That´s mainly mold like penicillium or aspergillus you see here,
probably brought in by yourself.
The blob is bacteria.

Maybe the little white dots near center could be cube myc, but too early to tell.
Also maybe there´s some in the top plate at 12 o´clock, but can´t see clearly.

I´d start some fresh plates and let these two do for some days,
even if it´s just for science.

Have a look here, it´s a lot to read, but also a lot to learn.
Especially for you the "working with spores" section and streaking techniques.
Mind your hand positioning while working with opened plates.

Thanks so much Goatrider - lots of information in that thread, I appreciate your introducing it to me. The wet cloth on the bottom of the SAB is a great idea. And sorry for the typo - I'm not sure what happened there. Mazatapec of course.

It has now been 5 days more and I am wondering if you might be able to help me identify whether I have anything I can transfer to a new dish? Today's photos attached.

I can't find any information on how best to identify the desired mycelium from anything else growing. Obviously the orange bits are bacterial. The dark green bit is mould. But most of the rest as you can see is sort of thick fluffy growth, which isn't the patterns of "reaching strands" that I might have expected. Although I appreciate that different strains of mushrooms may have different mycelium appearance.

Can you advise at all on how to tell the difference?
And do I have anything here that I can transplant? My spore prints weren't great so if I had to start again I would have to buy some new spores.

Well, i wouldn´t try to transfer from these.
Myc will be enmeshed with molds, especially the bottom plate.



I marked a region that could have been fine to transfer,
but you want to do it way earlier.
I guess the plates are fully overgrown with mold by now.
Just shoot up some new plates, do some more than just 2 :wink:

Do you have any tips on identifying mycelium? Obviously odd colours are moulds/bacteria but I am having difficulty finding general guidelines on identification. I don't want to have to keep bugging you as I feel very respectful of your time and help.

I will do as you said and start again, but I need to order some spores because the spore prints I took are very faint and I wonder if they are contaminated anyway. I will take more care when I take spore prints from my next batch.

I did try a transplant from the region you circled, and it is now in its 4th day on the new plate. Is THIS mazatapec mycelium...?

Sorry, but that culture is not clean.  If you look at the pattern of growth, it wanders back and forth growing back ontop of itself at times.  Healthy mycelium is very orderly and grows radially from the inoculation point.
