Mutated/sterile Fruits and DNA Sqeuencing
    #27778744 - 05/15/22 04:34 PM (2 years, 4 days ago)

I would like to get my presumed semilanceata fruits sequenced to confirm the
species. Some of the fruits are sterile and are looking quite different to the ones
that are producing spore.

Will using deformed/sterile fruits affect the DNA sequencing result? I assume it's
caused by environmental factors but have to ask just to make sure I'm not wasting
any money.

A couple photos to show the difference in appearance:

Looks more like a galerina semilanceata than a liberty cap, no conical shape. Different expressions of phenotypes won't effect DNA sequencing

Not Galerina since the gills are turning purple as the spores mature.

I agree, different expressions won't affect DNA barcoding results.  I put a penis envy sequence into Genbank, it matches all of the other cubensis sequences 100%.  Full genome sequencing would see some differences, but that's expensive and a whole lot more data analysis work.

Great, thank you both.

Alan, over in the hispanica thread you offered to help me with the analysis of the data and showing me how to put it into Genbank. I had a look at the AlvaLab
homepage and was wondering what services I have to order to obtain the data that is needed. Here is a screenshot of the services they offer.

DNA extraction, PCR, purification and sequencing - so should run 17.5 euros.
