First signs of life.
    #27772762 - 05/11/22 02:58 PM (2 years, 8 months ago)

I checked the bag yesterday, but didn't pick it up. Just shinned a light in the box to check around inoculation point and saw nothing so just closed the box up. I thought something would be there by now so began to question the syringe I got. But I was sitting here thinking to pick it up and take a look. I picked it up and on the back side, mycelium is forming .

One more day and it will be a week since inoculation so not getting my hopes just yet to ensure it's not contaminated.

Here's a pic of the bag. What do you guys think? Does it look clean?
https://

Still excited because I have an agar dish germinating and another bag inoculated.

looks super bacterial to me. you want myc to be a little softer looking & consistent, not creamy, wet & patchy like that. it sounds like you went spore syringe to grain directly, which has a pretty high failure rate. you can get lucky, but more often than not it gets bacterial like that. your agar plate is probably going to give you a much better success rate assuming you do your work in an SAB/flow hood! bags can be tricky in an SAB so you might have better luck using jars if you weren’t planning on it

You are right. Spore syringe to grain. Rather jumped the gun before I got my agar dishes. But I do have an agar dish inoculated. So, going to wait on that and do a couple transfers. And I have SAB. The room I use isn't very large and it's basically empty with no air moving at all. The only problem that creates is major sweating.

It's only been a week so it's good to be a bit positive about it like you mentioned, may get lucky. But unfortunately, I believe you are correct. Going to give it some time and keep an eye on it to see which way it goes. There is a bright side to this though if it does turn out to be a loss: I know the spore syringe has spores and, I know what my mistake was. Or what increased the contam rate. I ended up putting nearly 6cc's of solution in the bag. Less is more.

Thanks for the feedback. And really considering the jars. When I get to a strong, clean mycelium in a dish, I will add it to grain jars. I have some spawn bags, but they were premade.

oh yeah you don’t need 6cc of spores in there lol. i think your agar dishes will prove much better for you. premade stuff can be funky & like i said, bags are usually hard to do in a standard SAB. get some clean agar & prep your own grains & you should be golden. i’ve been loving wild bird seed lately. like you said, it’s all just a learning experience & you’ll only go up from here!

You are so right. I've read so much on hear about agar and grain preparation. I ordered a pressure cooker so will definitely be doing my own stuff from here on out. So far, my agar dish looks ok, with only a little condensation on the lid; I have it upside down.

I agree, the pre-made stuff is iffy. 50/50 chance I guess considering if the person was sterile or not. And even then, there is always the chance. Even if I do my own stuff, there is always that chance.

I have a pretty decent sized SAB, but it is tricky working inside of it with a bag. The dishes were a breeze compared to it. But yes indeed, we learn as we go, experiment with this and that until what works best. Thanks for the advice and your feedback.

"Have a nice trip".

any time friend (: experiment around with different teks & materials until you find stuff you like. i recommend trying out a few types of grain & seeing what you like best. i used to use oats but since found i like WBS much better. i’ve also gone through multiple types of agar containers. you learn a lot by trying new (reliable) methods

there’s always a chance for contamination, but at least prepping it yourself it becomes easier to narrow down where your problems lie. i def recommend using jars in the future unless you get a flow hood for bags. prepping your own stuff is really rewarding when you get it down

I have rye grain, but it was pre-sterilized so never know the quality. Looks ok: Not too dry, not too wet, but we'll have to see about that. I've also read that rye is rather tough to work with. The WBS has been sounding more inviting so may try that.

I agree. Doing it yourself can narrow down things when it comes to contamination. Also, there's no one to blame but yourself and takes the guess work out of who's to blame by not ordering pre-made stuff; you know on who's side the mistake was made.

In the end, you are right. Doing it yourself will be more rewarding and more enjoyable when it comes to enjoying the fruits of your work and time.

exactly!! plus it ends up being way cheaper in the long run to DIY, on top of consistency & control over your shit. agar is really easy to make once you do it once & get some hands on experience & grain prep is a breeze. i highly recommend WBS as a grain but a lot of people also love rye. if you want to try oats i’d recommend testing a small bag before committing to a 50lb bag. some people love them, some people hate them. i’m not much of a fan & i still have 30lb of the shit sitting unused in a closet now lol. WBS prep is super simple, just a 45 minute soak in some boiling water taken off the heat, let ‘em dry over night in the fridge or dry them for a few hours on a drying sheet/tray. they shake well & look nice once they get colonized & have looked much cleaner than oats for me.

Thats the truth. After all was said and done, I was up to about a $300 investment. That does include everything though. From spore syringes all the way down to the little things. Extra syringes, gloves and things like that.

You said it right with hands on experience. I have discovered that plastic petri dishes are very fragile. I set my SAB up, taking time to clean very well. Cleaned my agar dish, my syringe, flamed sterilized, placed everything in SAB. Was very slow and precise with my movements. Unwrapped my dish carefully and slowly, took syringe and placed 3 little drops at different places, then lid back on. Took my parafilm and was stretching it around and bam...the sucker cracked, lol. I have to be more gentle .

Anyway, I will have to look into the WBS; sounds very simple for sure. Rye may be good and suggested more as a favorite, but it's a bit pricey depending on where you look.

Depends on where you look. I use to buy mine at Walmart from $7 to $20 depending on the size. (I bird watch from time to time ).

But there may be a certain type you have to get. Not sure about the difference in like economy WBS or backyard WBS.

Rye on the other hand can go from $10 to almost $90 for a bucket. So it's a bit more pricey.

lmao yeah i feel ya, there’s definitely a learning curve with a lot of stuff in this hobby & a lot of it is weird stuff you don’t really see mentioned. learning first hand is the best experience

i think the brand i use is pennington? but anything that doesn’t have corn in it should be fine. mostly milo & millet is best

Ya there like $20 to $30 usd for an all in one bag that has some grain and some substrate on top. For a bag of just grain normally around $20. Definitely making a decent chunk of change if they can sell em.

woah that's a big margin, you can produce hundreds of these a week with a small gourmet steam sterilization setup
