These photos are of the first fruits from one of my APE/Ps. natalensis hybrids.

I mated monokaryons to create my cubensis/natalensis hybrids. I used 4 different cubensis varieties and a total of 144 pairings. It was greatly successful.

Here is a spore from these fruits that has germinated on my agar.
I took a swab and went directly from gills to agar. There's loads of germination and I have lots of candidates for F1. I will be approaching this slightly differently though as I will isolate monokaryotic cultures from these spores and pair them all together to look for recessive pairings to ensure albinism. After fruiting them all I will select an ideal candidate for F2.

Makes me want to pickup a microscope and experiment using electrical fields to hybridize species. I'm really interested in cross species hybridization. I really want to see a pan cyan /cube hybrid. I have a theory about bad genetics such as penis envy and how by correcting its genetics with a mutant / different species readily hybridize or with an electrical field would make it easier. I'm seriously thinking about making the leap! This thread rocks!!!

Ok ok, now hybridize the other two close relatives of cubes: psilocybe indica and psilocybe chuxiongensis. Then hybridize the results with your cube/nat hybrid. Spend the time to stabilize and....
You have invented: PSILOCYBE PANGEA

thats pretty rad dude, i remember posting about my own attempts to cross natal and ape by just mixing grains but you obviously went a faster and better way. Great work!!
What I did was clone this fruit, which I believe to be some kind of cross:

The white one actually turns into this, from a very APE pheno to something different. Seriously it changed colour/pigment.
shroomery creating a new species,? does this qualify?
how to measure beyond visual look...

I now have monokaryons of Ps. pseudoaztecorum and Ps. indica so I will do just that! Pairing them up in about a week and I'll let you know how it goes. I'm assuming they will be compatible.

I do not believe it is possible to cross dikaryotic strains in any compacity, including mixing grain together. What you need is monokaryons and then to confirm compatibility with clamp connections. Spontaneous dedikaryotization is very rare, and probably even more rare with cultures on favorable substrates like grain. But there's always exceptions and surprises. I wouldn't call that a hybrid because you just can't satisfy the burden of proof but it is certainly possible for cubensis and natalensis to hybridize, as I have shown.

What are the colonizing times like on these?
Are you growing them on straight coir, cvg, poo?
Do they need to be cased to help with blobs?
Last but not least, potency. =)
Very very cool project and I can't wait to see how it turns out.

Very very cool project and I can't wait to see how it turns out.
The colonizing times are very slow, almost twice as slow. Very much like APE.
And no there is no blobs or casing.
Yeah these are on straight cc/v.
And the potency is great, I really enjoyed the ride.
Thanks man! I'll post updates when I can.

I'd be interested in a break down of your process. Are there any threads detailing what you went through? If not perhaps you would care to write one some day? A picture guide on mating spores / strain crossing with a scope would get a lot of attention.
You've greatly piqued my interest - I'm into the idea of getting a microscope more than ever now. Which one do you have?

I will work on giving a visual outline in the future but for now I'll just give you a quick run down here. I germinated APE spores and isolated the monokaryons using my isolation technique, then my friend Yoshi Amano germinated Ps. natalensis spores and also isolated them using my technique. He handed me the natalensis monokaryons and I paired them with 16 of my cubensis monokaryons in some multiwell culture plates (48). I did a total of 144 pairings. After they had some time to colonize the wells I took small samples of the mycelium with a scalpel and checked the hyphae for clamp connections. Once I confirmed the presence of clamp connections I isolated the pairings in petris and then fruited them.

I'm glad you find it as fascinating as I do! I use a Amscope B120 compound microscope.

Yeah that's correct, this is a species hybrid! These fruits are the two species fused together and the offspring will be the new hybrid species. Haven't decided on a name for it yet.

I feel like I agree with the current classification that it is its own species, as it's much different than the current cubensis subspecies, in my opinion.

Thank you! I will post updates when I can. Hopefully sending some next generation cultures to grain next week.
I have more cubensis/natalensis hybrids in mind, and will be doing the pairings for that soon. Now that I have developed my dedikaryotization technique to include cubensis I have thousands of possibilities to work from. First in line next is Omni X natalensis.

I was starting to think you were Yoshi. I listened to all of the talks that he posted, cool stuff also, species is kind of a grey area. Different species can mate and produce fertile offspring. Savannah cats would be a good example of that, a hybrid of a serval and domestic cat. I read the F1,F2, and F3 males are often sterile though. So I don't see why nats wouldn't necessarily be there own species.

Yeah I'm not Yoshi, but he is my best myco friend lol. We work on basically everything together. And yeah he has been doing great things and I'm glad to see such a positive response from everyone. And yeah that is correct, it's definitely a grey area and there really is no telling what the results will be until it happens. I feel like natalensis is it's own species, it has enough differences for that to be the case. Lots of species in the psilocybe genus have very closely related counterparts.</font>

Typically when two species can be so easily crossed, it indicates that they're closely related enough as to be considered the same species.
Since Nats can cross so readily with cubes, as has been demonstrated by Judge_Doom, it is likely that Natalensis should be considered a "subspecies" of cubensis, which there has been some ongoing debate about since Natalensis's original classification.
Note that there is a difference between a subspecies, a variety, and a cultivar. Subspecies indicates a different, yet compatible, genetic line that arose, specifically, due to geographical isolation from other populations of the organism. Among subspecies there can be naturally occurring varieties (the examples are usually the Bengal Tiger, being a subspecies of Tigers, and the various colors of Bengal tigers each being different varieties within that subspecies.)
A cultivar is, of course, a variety that was bred into existence by humans (it's actually at the same level of subspecies in the hierarchy, usually, but it's sort of its own thing.)
In this case I think Judge_Doom has proven that this species should be reclassified as a subspecies of Ps. Cubensis with their breeding work.
Some cross-species breeding can be done with transgenic (genetic modification) techniques, but that's not what's going on here.
I have a question. What is it specificly that makes the conclusion if nats can cross with cubes, that makes it possibly a subspecies of cubes, or the same species?

That seems reasonable at first but there is a lot of fungal species that are capable of hybridization with another species. It's so common in the fungal kingdom that it kind of makes the grey area even more ambiguous.

Thank you! I have fruited a couple Melmak TP X Ps. natalensis hybrids but yes this technically is the first. Although I have more hybrids at this point with different species. Cubensis and natalensis were the first species I successfully hybridized.

Thank you very much! That's great to hear! I definitely plan on keeping it up as much as I can take on alone.
Those are great names 😆 I tossed around napetalensis for a little bit. Haven't decided on anything yet.
Alan hasn't confirmed the cross with sequencing but I was very thorough and precise in my methods so it's not really necessary. To my knowledge Jakeoncids hybrids haven't been sequenced either, at least not by Alan. Now I'm not saying there isn't any room for error, that's a given, but I am saying that I'm fairly confident. I have around 100 crosses now, maybe more.. not sure. When you're working with monokaryons you can generate dikaryons/crosses endlessly if you have access to some variety. I will probably have around 200 crosses by this time next year, maybe more. Depends on how stuck I get on stabilization.
Anyway, thanks for commenting man. Mush love

Well thanks! Here's a photo of some slants I made up to store the crosses, this is less than half of them.
I listened to that podcast! I have been trying to hybridize cubensis and ovoideocystidiata for awhile now with no success. I'm beginning to think you may need a forced method of hybridization to achieve it. I haven't started working with any protocols like that but I'm fairly confident it would work. So far I've been only doing pairings which are naturally compatible, like cyanescens X weraroa. Mush love from America

That's absolutely true, the morphology of the monokaryons is always very unique and atypical. You can definitely tell that each one is an individual.

My APE cultures looked like that, I think mine may have been an agar recipe issue tho, turned more rhizo once I put it to grain and spawned it. How do these hybrids compare in terms of effects? The nat effects seem to be more sought-after.

Interestingly the parent culture which produced the spores I used for the hybrids was a rhizomorphic culture. It goes back and forth though, agar recipe issues notoriously throw off morphology. And you know what I'm glad you asked, the effects are probably some of the best I've ever felt from taking something in that section. The trip was euphoric and didn't have any negative load on the mind or body. Really looking forward to the F1s to fruit so I can take more! The jars are almost colonized finally.

I absolutely agree. And nope, no issues with overlay. The cultures are tomentose though.

Thanks for posting this experiment. I've been wondering if they would cross being so genetically close. Might have to try this myself. I have a nice batch of Nats probably starting to drop spores right now lol. I also have PE x APEU F2 coming up as well. Sounds like a match made in heaven.

The APE X Ps. natalensis F1 'D' tea was very nice. Clean and low body load, but not as visual as I'd hoped. Similar to how the trip was for the parent hybrid.. Today I'm enjoying a nice afterglow, hope every has a good weekend!
APE X Ps. natalensis F1 'H' is maturing a first flush. Here's some quick pics! More to come.

I’d suggest definite back-crossing with the PE x APEU… I couldn’t get them to settle down even at F4

Maybe it’s all the PE genes lol. Lots of variation to choose from tho

Yes they do, thank you very much!
And of course, where the gills meet the stipe is one of the most important and beautiful features.
I give them currently as much FAE as I can, but I have plans to up it slightly, where I can. I'm not doing any tissue selection for these hybrids, only stabilizing through MS.

For anyone interested in a discussion on the topic of hybridization I was recently a guest on a podcast with a friend of mine to talk about various aspects surrounding the topic as well as giving a general rundown on how I created my hybrids.
Here it is in full!

Yes I am cloning it! Taking spores to agar too!
I'll have to try more fruits to answer your question, so far the trips seem consistent and in line with expectations.

Have you tried crossing nats and cubes with non monokaryotes? With simple more common methods?

No, I haven't. I only work with monokaryons when it comes to breeding and hybridization.. Wether I have to isolate monokaryons from MS or dedikaryotize a dikaryon I will end up with monokaryons.

Forgive my lack of knowledge on genetics, but would cubensis and ovoideocystidiata be able to hybridize?
I just remember Alan posting a phylogenetic tree of Psilocybe and I believe cubes and ovoids were more closely related than other woodlovers
Quote:
Alan Rockefeller said: Someone sent me a Psilocybe natalensis ITS sequence a few months back, so I had a pretty good idea what P. natalensis was - but it wasn't my sequence so I didn't want to share it. This week I made a Psilocybe natalensis sequence in my lab, which matches the other P. natalensis sequence I have. I've also uploaded it to Genbank, the first sequence of this species there.
The strain is Natal Super Strength.
Photos of the collection: https://mushroomobserver.org/472561
Here is a tree I made which includes P. natalensis and related species, with P. allenii as an outgroup.

The Psilocybe cf. cubensis sequence is interesting - it's quite close to cubensis, and has some noise, but I don't think it's just a bad read of cubensis because it has some of the same nucleotide substitutions that Psilocybe ovoideocystidiata has. Gaston Guzman collected it in Nepal in October of 2001. Without studying the collection (it's in XAL) I can't tell if it's a new species or if it's something which hasn't been sequenced yet like Psilocybe pseudoaztecorum.
Other things you can see in this tree are that Penis Envy has the same ITS sequence as regular cubensis, Psilocybe ovoideocystidiata from the east coast is the same as it is on the west coast and Psilocybe thaiaerugineomaculans is the same as Psilocybe thaiduplicatocystidiata.
In case you'd like to experiment with making phylogenetic trees or analyze the natalensis sequence yourself, here's the FASTA file: http://images.mushroomobserver.org/natalensis.fas

I have attempted to hybridize cubensis and ovoideocystidiata with no success but I believe with a forced method it is possible, probably not naturally though.

I just found this thread and I’m blown away. Very exciting to watch this unfold, and even more exciting to think you would release spores to the community when they’re stabilized.
I’d love to hear a more detailed trip report.

Thanks brother! Nothing I love more! 😁 I have thought about it but Yoshi was doing one for a little bit and I was a guest on the Myco Geeky podcast.. might end up doing that one again. I'd be happy to come on any other podcast though, it's a lot of fun! And great, you like woodlovers? I gotta get that one back out and try some stuff with it!
I have some spores of my APE X AA+ (AAPE+) cross left, DM me your address if you want a free set.

Any ideas of wtf they’ll resemble most? Or do you hope for a certain freak?
Do you have a thread for the werora cyan cross? I’m so intrigued I don’t want to miss that .?

That's exciting. Are these statements still accurate with the F2 fruits?

Yeah these are on straight cc/v.
And the potency is great, I really enjoyed the ride. ... The trip was euphoric and didn't have any negative load on the mind or body. Really looking forward to the F1s to fruit so I can take more! ... The APE X Ps. natalensis F1 'D' tea was very nice. Clean and low body load, but not as visual as I'd hoped. Similar to how the trip was for the parent hybrid.. Today I'm enjoying a nice afterglow, hope every has a good weekend! ... I've decided to name this hybrid species Psilocybe volanspateria.

And no there is no blobs or casing.
Yeah these are on straight cc/v.
And the potency is great, I really enjoyed the ride.
Correct! The phenotypes are staying prominent too. Thick and viscid caps like APE with bruising in the center of the pileus, yet it has the stipes and continuous growth/fruiting rate like NAT.

There is none, sorry. I was offering spores of an unreleased cube cross but ran out of those pretty quick. I'm not running anymore deals with the fundraiser until I release the Enigma cross which will be through the fundraiser.

This is the real judge doom, y’all. He’ll gladly take your money and help but not return the favor. Trust me, he thinks we’re all idiots
This is not a place for you to beg for equipment so you can grow products and charge ridiculous prices for swabs that cost you .16 and 10 seconds to take.

You don't know a single thing about me. Haven't you got anything better to do than bitch and moan about someone who is trying to accomplish their goals? I don't see any other cubensis hybrids around available, I think the problem you have with the price is a reflection of your own shallow emptiness. Grow up.
When you do something completely original I believe you're entitled to set any price you wish. Look at this whole thread and see the work involved. You seriously say that took like 10 seconds? Get the fuck out of here.

For what? And for what? He literally did nothing 🤣 all I did was state that the spores are available which is what multiple people on this thread were asking for.
I'm sorry you can't get a special deal on a release that's completely unrelated to the fundraiser. Did I ever elude that there would be some special deal for donators once I released the hybrid?

Do you see anyone else selling spores directly on the forums?

If you wanna sell overpriced spores you gotta pay to advertise like the rest of the vendors

If you feel you’ve been wronged take it up with a mod but faht has crossed and given away tons of spores and he’s not about you teasing then using the post as an ad.

So someone that’s admin on multiple mush groups has no idea what spam is and that websites have rules against pushing offsite vendors or how to contact other admins to become a vendor?
Come on man.

Look at my posts, obviously I don't use this site often. I'm on Facebook where it's simple. I'm just trying to share my work with people. I'm not trying to spam, I apologize that it seems that way. I never had a problem.
I felt that everyone here who watched the progress was entitled to an opportunity to grow it.
I don't spend a lot of time not working, so most of my posts are when I get a second to post. I'm too busy reading and working on these projects.

You guys can do whatever you want. Had I known it was a possibility to become a vendor I definitely would've pursued that. Had I known I was going to strike a nerve with everyone I would've avoided that.
But I can tell you that assumptions are not the way to common ground. We all have different strengths and weaknesses. I never wanted to hurt anyone.

Feel free to enter a Support ticket and plead your case to the admins to have your account reinstated. I’m fine with whatever they decide to do.
It just looks bad that you don’t come here often except to self promote and also looks bad to the current vendors we do have that pay for the ad space.. it’s not fair to them.

Doom, this is a dope thread and I’m very stoked to see this cool cross. And I totally support you selling these genetics, as people are going to love this. But this thread is not the place to do it. What I absolutely hate about using Facebook and Instagram is the constant bombardment of commercials and advertisements. I hate being treated as nothing more than a consumer and there’s no better way to feel like that than spend some time on social media. I’d probably be more active in those mushroom cult circles if I didn’t have to constantly wade through people shoving their products in my face.
The shroomery seems like one of the few places where I don’t feel bombarded by advertisements. Sure, there’s a couple banners around, but compared to most other websites nowadays, it’s relatively calm with ads. And the fact that faht shut down unauthorized vending so quick makes me that much more appreciative of this site. You’re also not the first one to get shut down from trying to sell stuff, and definitely not the last.
Like I said, sell those spores if ya want. People throughout history have made money of cool things they have made, and there’s nothing wrong with it. And you have some really cool genetics, so share them however you want. But obviously you know there’s a huge trading community here at the shroomery (and other places too). Spores are being given so freely here that I’m always surprised to hear people actually buy spores. I have yet to spend any money on spores/genetics since I bought my first ever spores, and that’s due to how generous the mycology community has been, mostly people I have met through this website.
You can totally do whatever you want with your spores and I hold no ill will towards that. Just don’t be surprised when you get shut down for breaking the rules.
