Panaeolus agar recipes
    #27722233 - 04/05/22 03:52 PM (2 years, 9 months ago)

I have had a bitch of a time getting any of the Panaeolus spp. spore syringes that I'm forced to buy (until I can trade) to produce viable mycelia on LME agar. P. tropicalis has worked, P. cyan. BVI and HI have grown, but cambos, bispora and about 5 more, nothing. The spores sit there and do nothing, no growth after two weeks. I find most syringes highly suspect and don't understand why these vendors don't sell more prints, but I'm also thinking that the agar recipe needs to be different for a lot of these. I have ordered some 781 plates from a reputable producer and will start making these as soon as I get up the courage to boil horseshit in my kitchen. What recipes do you use? Is this a big factor in getting many species to produce on agar?

It can take a while for spore solution to germinate.
Also depends on your technique,
did you streak out or just placed drops?
I usually rotate between malt and brf agar, no matter what species.
If you have trouble getting germination, try brf pucks.
Just shoot some liquid unto them,
and you should see germination soon, if there´s something viable.
Transfer with the poke to agar for further cleanup from there.
Syringes usually mean a bit of work, patience needed

I streaked all these plates with a sterilized loop.

How long should I wait before letting the vendor know their spores are shit? I've heard stories about spore syringes (assuming they're being collected/stored correctly) lasting for years, but I'm having a hard time buying it (in this case literally) when I've had so many contamination problems, given that I have a flow hood, have been working hard on technique, pouring agar, etc. Hundreds of bucks on spores just flying out of here and so many problems...

Dreaming of the day when I can participate in the trade system - that seems a lot more in keeping with the values that I associate with the endeavor of growing these species.

Regular light malt extract agar recipe works great for most species of mushrooms.  Including panaeolus/coplandia.

Other details I should mention is that there's no problem with contaminants with the spores that won't do anything, although some of the agar I used is Stamets' antibiotic LME...shouldn't affect anything, right?

Also, the Pan cyan HI is doing fine, growing reasonably well and I expect to have usable plates from this variety at least in a few weeks.

I thought maybe it was a temperature issue for the tropical species/varieties but the bisporus spores are also doing precisely nothing. What's the shelf life for pan spore syringes, usually?

I read in another forum that 781 agar was just the tits for this taxa. In my experience, this has been not the case. Has anybody actually used this recipe for Pans?
