I thought I’d start a thread to document my first attempt at growing, and leave a consolidated record of all the steps and lessons learned both for myself as I progress in my skills, and for other noobs who come along and want a one stop road map for the full process.

I started with a spore syringe from a trusted vendor, and decided on the plan I outlined in the noob forum based on tried and true methods.

On 1/29/22 I did the No pour agar with potato flakes and glad mini rounds. I made up 16 rounds of agar, drilled 1/4” holes, put micropore tape over the holes, put a folded up section of paper towel over the top, and wrapped the tops in foil. I put them in the pressure cooker and pasteurized them for 45 minutes at 15psi. I let them cool overnight in the pressure cooker. Only 15 survived the PC.







On 1/30/22 I moved the mini rounds into the SAB after cleaning the inside of the SAB with 70% Isopropyl, putting it face down on a towel in a small room, turned off the heat, sprayed the inside of the SAB with water with a little soap and moistened the towel with the same to catch and hold floating contaminates (next time use bleach solution on the towel?) and let things settle for 15 minutes then moved the mini rounds into the SAB, using gloves hands washed in alcohol (OB shoulder length gloves) then did each step for half the rounds at a time:

I pulled off the foil, wiped down the rounds with alcohol soaked paper towels, cracked the seal on the lids to make removing the lids with one hand easy, lifted each lid, squirted a couple drops with the syringe (which used a fresh sterile needle and was wiped with alcohol on entering the SAB) into each round, placed the lid back on the top. After all we’re inoculated I snapped the lids solidly closed, moved them to the edge of the SAB, and loaded and repeated the steps with the next 8 rounds.

After inoculating all 15, I moved them into the temperature controlled closet (77-80 degrees, no light) and waited taking a peek every 2 days.

2/5/22 3 of the rounds have what looks like the start of mycelium growing after 6 days. No signs of contamination in any of them.

Updated OP to post new pics.

Looking for advice:
I got germination in 3 of 15 agar plates after 2 weeks. No signs of contam in any as far as I can tell. (Any thoughts?)

I’m thinking about doing agar to grain with 2 of these three, and then trying grain to grain. With the third I’m going to try agar to agar.

I hated working in the SAB and got a 2x4 FFU this past week. Looking to get this set up to do the work. Dedicating a new small room in my house for my mycology experiments.

Here are the 3 plates growing mycelium (the multiple growths on a couple of them are a result of 3 drops coming out of the syringe, I believe):


2/18 As of today, 3 of my agar plates colonized with mycelium. Two of them fully, and one of them was about 3/4 covered as it got a later start. 12 of them never grew at all. I asked for some advice about reusing my failed agar plates. There was no sign of contamination, so I figured I’d just experiment. Using my new FFU, I was able to transfer mycelium from the 3/4 covered agar plate to the 12 dud plates. I will see if this gets me new growth.

As for the 2 fully colonized plates one is going to grain this weekend and I’ll try to do g2g when it colonizes, and one will go to Petri dishes as I’d like to try and pour agar into Petri dishes rather than use those glad mini rounds doing the no-pour technique.



2/19/22

Took the green agar that didn’t fully colonize and used it to inoculate the 12 agar plates that never germinated. First time working with FFU.

2/20/22

Checked this afternoon to see the mycelium starting to grow on all 12 agar plates. A lot of growth in under 24 hours.

2/28/22

11 of 12 of the re-inoculated agar plates took off. I have 13 no-pour agar plates ready to go to grain, and I prepped 10 quarts of wheat berries yesterday. Gonna make the transfer tomorrow. Excited that these all look contam free as far as I can tell. My sterile technique and new flow hood seem to have held up.

3/1/22

3/1/22 did my first transfer from agar to grain. 10 quart jars. Forgot to sterilize the scalpel for one. Used only parts of agar for 6 of the 10 taking the best looking sections since I didn’t isolate on my first grow attempt. Found 1 of 15 looked like possible contamination so I tossed it. Love working with an FFU instead of SAB.

Now let’s see what grows!



3/14/22

13 days since dropping the colonized agar in the grain jars. 9 of the 10 seem to be colonizing, but definitely at different rates. When I did the agar, only a few colonized from the initial inoculation, I then cut up one of them and inoculated the rest. The grain jar growing the fastest is from this second round of inoculated agar, and one where I used the whole thing. I also used a whole agar plate from the initial batch in 2 jars but those are growing slowly. I think the younger mycelium is a key here.

The remaining 7 jars were inoculated from the best looking wedges I could find from the remaining plates. All but 1 of the 10 grain jars are colonizing. I’m not seeing any contamination. I’m inoculating at 72 degrees (and even heating just a small closet to that temperature is costing over $2/day). I have an old mini fridge that I am going to sacrifice and make into an incubator for Petri dishes and grain jars. I’m going to grow the mushrooms in mono tubs in a cool house, and trade off speed for denser and hopefully more potent fruits. My lab is right around 58 degrees most of the time. I can put the mushrooms in my living space and let them get light and heat on a natural day/night cycle and they would be 63 at night, 68 by day. I’ll probably do this. It’ll be warmer in the summer, but air conditioned.

3/24/22
I have one grain jar that is taking off much quicker than the others. 9 of yhe 10 seem to be spawning mycelium in the grain, and there is no signs of contamination that I can see. I shook up the heavily colonized jar and distributed the mycelium through the grain today. The collage of the 4 jars is the same jar from all 4 sides. The other picture shows how much slower the other jars are going.



3/28/22

Well, 4 days ago I shook my best jar. Looks like it wasn’t a mistake. I could see that the mycelium had formed a big ball in the grain, it wasn’t just on the surface. As a result, I decided to shake the other 9 jars. 7 were slow growing but looked good. One looked like it may not grow, and one looks possibly contaminated. We’ll see.

In the mean time, here the shaken jar 4 days later. Looks great to me.



3/30/22

Well, my good jar was mostly colonized and I gave it one more shake. Not sure if that was a mistake. I’ve read competing advice about how often and how many times to shake. My slow growing jars all got their first shakes yesterday. It’ll be interesting to see how many fully colonize. A couple look like they’re already taking off.

4/1/22

Gave jar 3 a second shake today. It was catching up with jar 1. I think next weekend I will do g2g with jars 1 and 3 and multiply them to 10 jars.

4/2/22

Shook jars 5 and 8 today to see if they can catch jars 1 and 3.

On 4/3 I put jar 1 in the fridge. On 4/5 I put jar 3 in the fridge. Waiting for jars 5 and 8 to catch up.

Shaking jars 4, 6, 7, 9, 10
Jar 2 which had a small agar wedge that I thought was not viable is showing first signs of colonization.

4/7/22
I have decided to try Dino soil vs Cvg for my bulk grows. I ordered a 10pound bag of Dino soil that will be here on the 12th. As soon as all the jars catch up, I’ll spawn them to 4 tubs. 2 to dinosoil, 2 to CVG. These 4 tubs will help me figure out if Dino soil is worth the expense for ease of use and quality of fruit. The CVG based on what I already spent will be just under $3/tub. The Dino soil at this small a volume would be $9.56/tub, but be pasteurized and hydrated already, saving a ton of work in the kitchen, especially cleanup. If I buy 100 pound orders in the future, it comes down to 6.25/tub. My mini tubs should require 2.5 pounds each. So a little over twice the cost of making my own CVG, but with extra benefits that may be worth it.

4/9/22

Jar 2, which had minimal growth (maybe 20%) finally taking, and jars 4,6,9,10 which were well underway, were all given a shake.

4/11/22

Pulled jars 1 and 3 out of the fridge today. Plan to go to bulk in a couple days. Have 5 jars looking ready. 4 are catching up. One looks like it may be a lost cause. We will see.



4/15/22

Did my first spawn to bulk today. 5 colonized quarts into 5 shoeboxes using the SFF Shoebox TEK. 1.5 quarts dinosoil per quart of grain. 1 quart mixed with the grain, and 1 pint on top as a pseudo-casing, misted, and then the lids were latched closed. I’ll be doing the other 5 quarts soon with CVG as soon as the last jar catches up for that batch.

The jars smelled fantastic when I opened them. I was unsure what to expect, but the aroma was quite delightful. I’m guessing no contamination in the jars based on what I could see and smell. Let’s hope they grow!



4/18/22

2.5 days since going to bulk. Shoeboxes seem to be colonizing, humidity levels in the boxes seem ok. Fingers crossed this is mycelium and not some kind of contam. Anxious but optimistic and excited.



4/22

7 days after going to bulk. 4 views of a typical shoebox.

House is 63 at night, 68 by day.



4/29/22

2 weeks since going to bulk. Looks like most of the shoeboxes are closing in on full colonization. Surface conditions looking good. Sides and bottoms are 95%+ colonized. Tops are 85%-90% colonized. I’m growing in cool conditions (63-68) and surface conditions look good with tiny dew drops on the surfaces. Hopefully I hit full colonization within the next week.

4/30/22

Got my first pins!

Surface conditions look good in all 5 shoeboxes. One of them is showing pins! Real live visible pins with no ambiguity. $2500 in equipment, months of reading, months of working, and I could maybe be growing a mushroom!



5/2/22 many new pins coming up in the tub that pinning. No pins in the other tubs yet. Anxiously looking forward to seeing some. Surfaces all have tiny beads of water. Hopefully there will be some pins in other boxes soon. None the less, the fact that I’ve gone from spores -> agar -> grain -> bulk -> pins on my first grow, with no signs of any stage failing (and having learned a lot in the process) makes me very excited about my next grows.

5/2/22 - night time update

A second shoe box has begun pinning. The original shoebox to pin is showing many new pins, and the originals are growing fast. A couple shoeboxes look like they have some primordia or hyphal knots forming. Staying hopeful for 5 good harvests.

5/5/22
Based on advice from the forum, and most importantly a TC, I opened the tubs and misted today. All 5 tubs had at least a couple small pins, and one tip is coming in. I’m not sure why one was so much more prolific. It wasn’t my best jar, they all had the same substrate, ratio and pseudo-casing. The pseudo casings were all misted identically. Whatever happens at this point, I have a nice cluster to clone, and my first grow is yielding mycelium and fruit and not contams, so it’s a success no matter how small the yield. I learned a lot. Next round will be better.

5/9/22

So today I harvested my first flush. Although all 5 shoeboxes were prepped identically, only 1 really took off. I think my substrate was too dry in general. I harvested all 5 boxes and 90%+ of the mushrooms came from the same box. I got almost 200g wet from 1qt spawn in the good box, and just 30g total in the other 4 combined. I harvested it all. I put 204 in the dehydrator set for 165 and 24 hours, and saved 26g of the baby mushrooms and pins that I plan to make tea from tonight rather than throw them away.

I also am soaking the subs for 3 hours based on the SFF factory shoebox tek. I harvested even the pathetic shoeboxes hoping this new moisture would give them some life in the next flush.

No signs of contams anywhere at any point in the process. Even though I’m disappointed in the yield, I’m thrilled with the success of my first grow, and how much I’ve learned for the next one.



5/15 - 5/22

Harvested mature fruits from second flush for a week. At the end of the week had 262g wet and 37g dried (165 deg for 24.5 hours). 4 tubs of my second flush are still growing and have a lot of pins and a few mushrooms getting close to start another weeklong harvest.

6/9

Last harvest was last night. In the end I got 5 good flushes from the dinosoil with big huge fruit totaling 1766g.

I got 10 flushes of small thin stapes fruits from the coir only boxes but only got 1369g from it.

Final lessons learned: dinosoil was not at field capacity. Required more misting. Will hydrate before trying it again.

Total yield for first grow: 3135g. Or just over 110oz. 8.9oz dried. 0.89 dried ounces per quart of colonized grain.

Overall I’ve learned a ton, enjoyed the journey, had some good trips, and got a pretty good result for a first time grower.

Next grow will be FP+ followed by nats. Many other varieties are on agar, but I’m not sure how viable many of my agar plates are. I fear I poured them too shallow and things are not going well. The FP+ is ready for grain, and the Nats are growing out well. So I have a few exciting grows to look forward to in the near future.

I only have a couple suggestions.

Next time flame sterilize your needle.

Also I'd make a homemade inoculation loop out of a twist tie on bread or garbage bags and use that to spread one drop of liquid over the plate. 

I do a Z pattern.

It might work fine the way you did it.  If not try these things on your next go round.

My plates get 12/12 lighting just like my tubs.  No need to keep them in the dark.

Definitely going to flame sterilize my needle next use. I though with a sealed sterile needle I’d be fine for first use.

Love the loop suggestion. I’m hoping to do some new plates tomorrow or the next few days. 

I really hate working in a SAB, and got a FFU to work in front of. Going to try using that this week. I have more empty rounds and plenty of agar.

I’m thinking maybe the other agar plates haven’t started growing yet, not because they failed, but because it’s only been 6 days. Still holding out hope. Worse case I learn to do agar to agar in the next week or so.

They’re only in the dark because I didn’t think they needed light at this stage and I haven’t set up lights in the closet yet.

Today 2 of the agar plates became fully colonized. I have 12 agar plates that never germinated. Can I do agar to agar in these 12 ungerminated plates? There is no contamination growing after 16 days. Should I do fresh agar? Also I can’t do it until Saturday. Too busy. Will the fully colonized plates be fine at room temps until then?

For sure do fresh agar. I know its a costly amount of your time. But hey, it's a hobby after all. I'd personally not risk it. Going to need fresh plates anyway. You have a LOT 9f transferring to do. Multiply it by at least three.

I didn’t get this advice before I went and reused the existing agar. I used the smallest mycelium colony I had and inoculated the agar that failed to germinate. I still have 2 full plates. I’m going to do some grain prep today and do 1 jar of rye berries tomorrow, and I’ll do some new agar and do some Petri dish pours as well.

The best part is that overnight the mycelium I transplanted to the agar that never took off is all growing great, and I got to use the new FFU for the first time.

It was all the time I had yesterday, took about an hour to clean the area, do the transfers, clean up afterwards. Tomorrow I’ll have more time. Prepping the rye and agar today and will leave the rye in the PC overnight to cool, then I’ll PC the agar tomorrow shortly before I’m ready to pour.

I just added the pics. This is what they looked like coming out of the PC. A little condensation but the grain is loose and not sticking and only a few popped berries.

So I’m thinking of taking the fast growing jar and going G2G and inoculating 5 jars. Grow the 5 jars out and go G2G and inoculate 25 jars. Hopefully grow all 25 and go to 5 mono tubs. This will result in way more mushrooms than I could ever use, but I have a lab grade freezer, a whole house backup generator in case of power failure,  and capsule machine. I could dry them, powder them, capsulize them and freeze them. They should keep for years based on what I read. I have everything I need in the house to do this big a grow except an additional bag of grain, and some more bulk substrate. Seems like a small expense to try and maximize this batch.

Can you clarify your takeaways regarding agar quantity and age during inoculation?  I'm also leaning towards cooler temps since I can't seem to shake this bacillus curse.

Based on my fastest growing jar, if I were to do another batch on No pour agar from spores I would do this:

1. Inoculate some agar (I only got 3 of 16 to grow, next time I’d drag across an inoculation loop to spread the drops in a Z pattern)
2. Grow it out and transfer the fast growing healthy edge to new agar.
3. Repeat transferring fast growing agar edges to new agar plates to multiply the fast growing sections
4. When I have 10 fast growing plates, transfer to 10 grain jars. I’d use the whole agar, not just a wedge.

I think my lack of contams is that I am really diligent about sterile procedure. I watched RogerRabbit’s videos (get these if you don’t have them) and I do what he does.

I wipe down my work area with alcohol and run the FFU for a couple hours before I start.

If working with a SAB, I spray down the inside of the box with water with a tiny bit of soap and work on a towel on my work surface that I spray down with a light bleach solution. I let the air settle for a couple hours before I start.

Block all heat ducts in the working room.

Shower, put on fresh scrubs, Tyvek sleeves, gloves, mask, sanitize constantly with spray bottle and alcohol. Flame sterilize scalpel every time it touches a surface.

I follow RR’s tips for holding things and moving lids.

I work really hard to keep contams out.

I did my first agar in a SAB and incubated in a much warmer closet. No sign of contams, so I think I’m doing pretty good with sterile process.

I decided to grow slower and cooler only because the cost of heating the closet, and I have enough dried mushrooms on hand to last many months for my microdosing schedule.

This is my first batch. Maybe I’m just getting lucky. I really am working hard to keep things contam free though.

Updated the OP with todays progress.

A question for those who might have some advice:

Should I have shaken up the jar today? Later? Sooner? What about the slower growing jars?
