Should I let Agar fully colonize before a grain transfer?
    #27629904 - 01/23/22 05:18 PM (2 years, 2 months ago)

I just got my first mycelium culture on Agar.

Should I transfer it to grain right away or let it fully colonize the Agar?

My theory is that if I let it fully colonize, there will be less chance of contam since there is nowhere for other life forms to hide. Is this true?

Contamination can live in, on, under and next to mycelium. Make sure you have a clean culture by transferring enough times to be sure it's clean. I usually let my plates grow out about 2/3 the size of the plate and cut pizza slices out to drop into my grain jars, but I don't transfer the edge piece of the plate into grains because I feel like the very edge is where any sneaky contams will be hiding on the plate.

Cultures can become contaminated even if they're fully colonized and it can actually be worse when that happens because they generally look normal. There's no point waiting if you want to use it now.

I agree with Kizzle, there is no point in waiting to use agar if you see the growth you want already

the only reason I let my plates grow out is so I can put one plate to 5 or 6 jars and be able to put a decent size wedge of agar per jar. Otherwise, if it's clean and ready then run with it.

here's a T1 ghost plate that will inoculate 6 jars tomorrow. The only thing left in that plate will be ~1/4" ring around the edge and the center transfer piece
