orange stuff in lc+annoying noob questions

I made a couple batches of lcs recently. One of them has this orange stuff near the needle. I cant tell if its dry myc or some type of contam. The other batch is all just white myc and some malt water.

When making my lcs, i never fill the needle to the top with liquid, i always finish by aspirating sone sterile air. this is to make sure that the needle doesnt contain liquid that could "wick in" anything. I also flame the needle from the base up before putting on the needle cover. Is this good practice? If not, how does one ensure that syringes remain clean?

definitely contamination IMHO

can't tell from the pics but I bet the LC looks real cloudy in the light, should be clear as clear except for the myc

that orange is all bad

Thats annoying, but part of the game i suppose. Any idea why it happened only near the needle?

When you aspirated sterile air what did you do?
Was this done using a flowhood?

Your syringes seem a bit opaque, notice how clear this lc is?

Thats what my jar looked like as well. The liquid could very well have been glass.
And yes, i used a flow hood.
Basically, i filled the needle with liquid to 90-95% of its capacity, remove it from the liquid, suck up some air until i see bubbles enter the lc, that way im sure that my needle is empty. Then i blowtorch the needle, from the base to the tip, till it glows. This sometimes forces out a small amount of liquid leftover in the needle, and ensures that it is totally dry inside. Then i add the cap.

From a physics standpoint, I don't think you need to suck in the air. Flaming the needle will turn the liquid in the need to vapor and clear it out.

I'm a total noob but this is immediately what I was thinking too. Good news if the LC s clear you've got the issue nailed down! I'm not up to making LCs yet but closing in! Good thread!

It was indeed pretty clear, so im guessing the issue came when making syringes. At least i have one part of it nailed down though.
How do you guys store them? Is the needle cap enough "protection" or do we need anything extra?

I put them in a fresh ziplock and in the fridge

I have seen people wrapping the lid at the neck of the syringe like a petri

and I agree when you flame the needle anything there is evaporated without the added risk of pulling in air
