Update: 3/27/22 original microwave jar
This was the original 3 minute microwave jar that I let sit for a month to see if any contamination would grow with it being un inoculated.
Nothing ever grew so I wanted to then put good mycelium on it to test that the grain was still viable and could host life on it so I transferred a wedge of summer white oyster in to it
The results speak for themselves,

I’m actually very suspicious of it, I think it might be good fungi mycelium some how, and that made me think, I grow a lot of oyster in that room, what are the chances the spore load is so heavy some oyster spores could have found their way into the jar through the micropore??

Funniest shit ever just wanted to give y’all an update The 2 jars that originally contaminated we’re actually contaminated by the heavy spore load in the same room I grow in lmao I’ve never heard of or Sean this but apparently blue oyster spores are able to get through micro pore tape , to my surprise I went to clean out the lab and forgot about those contaminated jars and sure as shit it was pinning in vitro

Also kept the 3 minute test microwave grain on agar around and looked at it yesterday, it’s still clean lmao 😂 didn’t take pics but I still can if y’all want

Oyster spore load is very heavy as you're aware, they definitely get through mp tape. I always gre my oysters in their own filtered tunnel.

not trying to dirail the thread but since there is a lot of slack time due the subject in hand I'm gonna go ahead and ask do you guys go back to spores after some time or do you just buy new LC from someone else? I'm asking because I'm growing supermarket clones and I'm not sure how long I can keep pushing it and if you do what is the best approach for isolation, i see a lot talking about cubes should I just select the fastest growing sectors? thanks in advance

I haven’t had to restart mine yet but what I like to do is make 10 slants when I first get the mother culture and then put them in the fridge so when you start to see genetic degradation, just start back at square one with a slant, depending on how much you produce that should last a few years before you would need to go back to spores

Yea it blew my mind that they where able to get through the mp tape, I guess it’s a numbers game, if the air is heavily saturated it just a matter of time before the dam breaks metaphorically speaking

Oh and I think for my next experiments I’m going try simmering grain with lime + oxyclean since it’s essentially concentrated hydrogen peroxide, and yes I’ll be using the “free and clear” version since it doesn’t contain any dyes or fragrances and the best part is when its heated it breaks down into oxygen and water so it shouldn’t leave behind anything that will hinder the good mycelium, and yes I’ve done research into concentrated peroxide being explosive, but this is still diluted enough that is not an issue, people actually boil deer skulls in oxyclean on the stove and they don’t blow up lmao

How about tannins? I once tried infusing grains with triphala tea. Triphala is an indian medicine made from three very tannic fruits. I made super strong tea from it, which would be bitter if anyone drank it, and seeped it into the grains. Nothing at all germinated in those jars, even though I added spores. Then I tried to open and close them a few times. And nothing still germinated in them. With a certain tannin content, nothing could grow even with all that available nutrition.
If one could infuse the right percent of tannins, one might be able to inhibit growth and not stop it entirely.
There are much cheaper ways to get tannins than special indian medicine - leeching acorns and sawdust would work.

I think wood loving strains would probably benefit the most from using tannins since they are probably already somewhat tolerant

A lot has changed since i started experimenting with lime but i have a few updates i want to share
I was able to successfully sterilize oats using atmospheric steam and lime for 24 hours
I’m planning to conduct more experiments with shorter time and i hope to be able to get it down to just an hour or two using atmospheric steam and whole oats
