If you took your spores from a single spore print does that still count as 'multi-spore'? In other words- is there genetic variation among spores from a single fruit?

I'm no expert but pretty sure that's right, spores all have different genetic variations even from a single fruit.

My jars are 1/2 quart and use one 1/4" hole for GE, seems to work ok. Means I can just stick on a piece of tyvek/micropore tape or polyfill it. If it's optimal, no idea. Some say 4 holes 1/8" and a SFD.

With this grow I was going the viable mycelium route in the need shrokms now mode so to speak.  But that being said when I transfer from multispore in search of a monoculture, I'll generally not see uniform growth untill T4 or 5 sometimes.

And I also made the brf jars completely a multispore grow because I previously made a liquid culture directly from spore.  I wish people on here would talk more about liquid culture for starring spores.  It's good if you test it on agar first and it passes.

Here's more photos of my brf jars chugging along.  I stated these were pint jars but they are actually standard pf tek half pints.

These jars are less than a month old.    Not sure if smaller jars can be harvested sooner than 2 months.

What do yall think about that??

So to clarify I took a spore print of p.
galindoi and scraped spores into my liquid culture.  After about 2 weeks I had good growth so I put it to agar.  Nice and clean so I ran it on these brf jars.    So yeah this is a mutlispore grow all the way.

Even the bags I used the lc.    But a few of the rgs jars were straight agar wedges and they are slower to grow than the lc.

Here's the lc

And here's more jars

quick question, are stones really dense? (i.e., high weight, low volume)

I haven't harvested any of my stone-producing jars yet but will do soon when I get back home in a week or so. I have a bunch of fully colonized Galindoi jars with many stones, I plan on trying to grow some fruits because they are very pretty and I'd like to secure at least a print or a few swabs.

I'm also not sure if I should put the stones in the substrate to let them get bigger or speed up colonization.

thanks for all the help and insight guys, love ya all :mushroom2::heart:

Just spawned these galindoi to bulk.  Can't wait for fruits and prints.  Then my grow will be a success.

Simple shoebox tek

Before

https://files.shroomery.org/files/21-47/797913134-20211126_152024.jpg

After

https://files.shroomery.org/files/21-47/797913271-20211126_153827.jpg

The jar I spawned from was one that was only 3 weeks old and I found a few tiny stones in it as well.

I'm more interested in the fruits so I'll let this consolidate and then maybe I'll case it but I'll most likely induce fruiting soon.

I've said it before and I'll say it again, nice bottles man do you wrap them pre or post PC? I'm transitioning to all glass widemouth pints like muda said he liked with unmodded plastic lids.

I don't really understand the question, but this might help (maybe).

I have noticed that stones grown invitro are denser than spawned. When I dry invitro stones, they are 1/3 of their wet weight. When I dry spawned stones, they are less than that.

If stones are left in the substrate, they'll grow bigger.

Awesome! I'm glad you had a good time :dancer:

I'll write a proper trip report, and tell everything about it: the when-why-how. I'll put a link here, and also PM you when it's up. It should be done within the next couple of weeks, also because I try to write down all my big trips into my journal, and I haven't done that one yet.

Been trying to get tampanensis started again from a print I took.

Kept getting aerial myc and thinking it was mold...

Put some to a brown rice/horse poo mix 4 weeks ago. Last jar not shaken.

Going to straight case with coir/verm and then immediate into fruiting.

Also interested if they can fruit without casing going into a JCM/Angam style chamber.

Still got a Rancho Agosto I've been meaning to get started, hopefully over the festive season.

I leave the lids loose and tape them in place with micropore before the pc and then after inoculation. So they are "filtered" from sterilization to full colonization.

I do wrap them in tin foil for the PC cycle but it's probably unnecessary

This is the final harvest of a mycobag Unicorn Type 3 TL of "Mex. A".

It was a pain in the ass to harvest because that clone produced small stones, even when spawned... :facepalm3:
Never using that one again

First half and second half of the bag (65gr are missing from the second photo, because I harvested them later). I had to spawn it to 2 shoeboxes:

Stones are smaaaall:

Before and after drying:

Final dry weight:

Some packed individual doses for later use:

Harvest numbers:
  • First shoebox: 369 grams wet.
  • Second shoebox: 418 grams wet (had an extra week on the 1st one before getting trich).
  • Total wet: 787 grams.
  • Dried down to: 228 grams.

Spawned stones contain more water than invitro stones.

When I dry invitro stones, dry material in equivalent to a third of wet material. Here it would have been 262 grams dried instead of 228 grams.
BUT! I'd never get 787 grams of wet stones from 6-7 jars, which is how many jars I can get into a mycobag.

Spawning stones rock!

Nice melgo.
I just spawned two more shoe boxes will harvest when they are fat and juicy.

The ones i spawned on the 16th of November already have stones ripping thru the surface!

I have a shoebox thats 3 months old. And never grew a fruit 😔.
But hella stones.

I wanted to share the agar recipe I use for sclerotias with you guys, because so far it has been producing stones very frequently for me with "Alt#7", "Mex .A" & lately "Rancho Agosto" from Ambrose (3 stones forming on agar, ~4 weeks after inoculating it with a spore print). The later is what made me think I should really share it here.

Agar recipe for sclerotia producing species:
  • 200 ml of water
  • 4 gr of agar-agar
  • 3 drops of food coloring
  • 1 drop of honey
  • 12 ml of instant purée potato flakes (2 level teaspoons)
  • 12 ml of coffee (2 level teaspoons)

It has already been said in this topic by I-don't-remember-who (sorry) that PDA is a good one for sclerotias (I'd been using PDA all this time by sheer luck), but I first read from RR that adding a bit of coffee to the agar stimulates stone production on it.

Hope some of you find this useful!

What is the point of encouraging stones on the agar? Are you transferring/cloning the stones?

I noticed aint getting shit for stones on old plates with MEA+ac and was about to put my cultures to grain soon.

The little experience I've had growing stones. My best producer was a plate stone clone. My guess is that it helps you get a more stone leaning than fruit leaning culture.

Stones on agar are usually a sign for a good stone producing culture. Yes I do transfer/clone the stone to another agar round, and then once grown out use that one to inoculate jars and/or make LC.

Just posting this here to “bookmark” this thread for the next time I grow stone producers.

Pictured - Psilocybe Tampanensis

I think the print I got was labeled ATL#7, but not too long ago the mycologists determined that ATL#7 is actually identical to Ps. Tampanensis
