I was making shitty low nute trash for a while alongside massive contam vectors. I fixed the vectors and moved to a more standard agar recipe. 10g agar, 8g malt, 2g yeast flake, 500ml distilled water. My cultures love it. It's a nice yellow/tan color and the white myc stands out distict enough I don't need more color. I could try charcoal again just to use it up.

Just getting over the flu so cult has caught up. Clear is superior. Black looks nice though. Been messing with master's mix/verm cake. No grain Some gals, pan cyans, enoki, shiitake. Semp about to hit bulk (by feel hay, coffee grounds, coir) Excited to dip into these dime bags of melted agar from over the pond. (solidified in fridge)

I just mix agar and malt exract 50/50. 40 grams of that mix into a liter of water and then sterilzed for 30 min. Plates arent clear put not opaque. I get strong growth and can see what i need to see. I've used coloring in the past and it does have its uses. I just dont use it much these days.

I usually do roughly 7.5g of both lme and agar with 500 mL water in a quart ziploc and hit it with the stick blender until it foams up to the top then pour and pc

Do you ever get a bad batch of plates? By bad I mean unsterile right out of the sleeve?
I ordered some cheap 100mm plates and roughly 50% had bacteria or mold within days. I chalked it up to poor technique and then poured multiple sleeves of 90mm from another batch, not a contam in sight.
Then as a test, I poured half 90mm and half 100mm from the suspicious batch in the same session. The 90s were fine and half the 100s contamed again.
I’m now confident that batch of plates were shit, and fortunately I only ordered a small amount of them, but it makes me hesitant about pulling the trigger on 500 dishes for something like that to happen again.

Sleeves of improperly sterilized plates is why i switched to glass plates and sterilizing them myself.... so, yeah....

Yes... that's what I'm coming to realize.. Even if I take the smallest piece of a sector like single hyphae.. I still see some genetic diversity in the transfer ?

Or maybe when starting with monokaryotic cultures from single spores for cross breeding purposes?

All this talk of nutritional yeast extract has peaked a question in my mind. How is myc with salt? The reason I ask is has anyone ever tried marmite as a yeast extract? Its concentrated yeast extract with salt added that you spread on toast. And 100% water-soluble.

Looking good. I can’t wait to get my hands on some nats.
Some PE T1s finally showing some organization. Check out that little tomentose sector at 2o’clock. I’m finally getting some vigorous growth. Hoping to go to rye after another transfer or two.

People will dilute spores and then use a microscope and a needle to scoop up a single spore and put it on a plate. Then it’ll grows but won’t fruit unless you introduce it to a second single spore colony and I think that’s the only way to do it.
But also there’s no point really unless you’re going to put tons of work into it. Narrowing isn’t always a good thing.

I would go really wide and then select clones before going narrow.
I tried dragging a caps gills against a few plates today instead of swabbing.
