Not necessarily.. that’s just what my reasoning tells me. I’m open to hearing what y’all think about it
Quote:
Scrimshaw said: I’ll take my chances moving a wedge from a sealed dish to a fresh jar 3 inches away before rinsing it in water first. Seems more complicated for no benefit.
If something could land on the wedge from the time the dish was open to when it went to the jar, why couldn’t something land on it after it came out of the water?
I guess I was meaning more like something getting on it while it’s in the dish during colonizing. I just hear ppl say that “the culture appeared clean when I added it to the jar” but then the jar turns to shit. I suppose the water could make the wedge more sticky to things in the air during the transfer.. maybe move the sterile water jar right next to the grain jar so there’s not even 3”.
But like I said, my reasoning could be flawed.. that’s why I’d like to hear what y’all have to say about it
Edit: if you’re usung SAB, I guess you want to be as fast as possible and adding that step would be counterproductive.. with using the oven bag, I have a bit more time to fiddle around and rinse tissue. If you’re using a hood, there wouldn’t be a point since it’s all clean anyway.. so I guess nvm lol

Hey guys, need a little advice as to whats going on here..
These are four days after first transfer from a very underdeveloped plate that I tried saving from penicillin. I'm not really sure what they're doing just yet and truthfully I can't be certain that last pic is even mushroom mycelium haha
And there's this Albino Blue Magnolia, looks like it had some hidden contam (bacteria?) that is taking to the plate before the ABM mycelium
Any advice appreciated

@the sterile water
Water is an amazing solvent. It can't hurt anything if you got a good technique. Especially for those that don't follow the crowd.
Faht uses a surgical technique. Did you come across that on your own? Or watch a surgery with 2 ports to work? Fuck it time to get graphic!
edit: it's 4 am I can't figure format. Fucking youtube. Google 2 port laparoscopic or 2 port pars plana...

Naw that sterile water is just one big unnecessary step. You'll find that the people that say their culture "looked" clean and fucked up when it went to jars are either people that don't know what a truly clean culture looks like, or they're still a beginner and need to work on their sterile technique.
With all respect in no way should there need to be this additional step. Nobody has ever felt it necessary, and for good reason, especially those knowing what they're doing and maintaining a 0% contam rate throughout.

When there are absolutely NO sectors in the mycelium. You will never ever achieve it from transfers alone.

My Ps. cyan culture.
This is T7. I'm just transferring to see how the mycelium changes over time.
I have a better candidate for these tests so maybe this culture will get retired.

Never as in it's not possible or never as in it's unfeasible?

Expensive yeast extract just seems expensive. I toss like 2g of flake and just shake a few times hot. The neon colors look nice, make some bright ice cream.

so many colors...
Didn't really do my homework as well as I usually do on the yeast. Saw a few mention it alongside clear and went for it -- flakes will be tested as well for science (and because I'll be doing this awhile and like to save $).
