I'm gonna inoculate like 30 bags today, 9 are not presealed, but I'm gonna wipe em down all the same.

I run my FFU constantly, and I always wipe.

I am not trying to argue wether to wipe or not, i am just sick of the amount of alcohol I use and I kind of feel that wiping with bleachy water would essentially do the same thing as wiping with iso. I still use plenty of iso for surfaces and other stuff, but I just can't deal with the smell anymore when mass inoculating bags.
I think iso is overused and only gives us a false sense of security and maybe just wiping the floaters is just as good as killing (99%) of them.
In the next weeks i will start to run some experiments and see whether iso really makes that difference. I thought I ask first what the current consensus is.

iso evaporates so u wont be sealing a wet bag which doesnt always work so well

I agree about fast evaporation being nice. And it's definitely not the consensus, but I use ethanol-based sanitizer so I can inhale as much as I like.

People who do lots of bags:

So I am going through lots of bags, I pre-seal, PC and set on the shelf in the lab and inoc them when I have 100-200. They don't really get dirty (as it's a clean room with flowhood always running) but I always like to iso spray the top of the bags and wipe before I cut it to sanitise them. You can imagine the fume this process creates after you do like 100 bags.

SO not only this is very wasteful and potentially dangerous, but I think it's completely overkill.
As an alternative, I  was thinking about a bucket with 10% bleach water (tbh soapy water would work too)  and dip the bags in for like a second to wash off any floaters. Does anyone do this?
What do you guys do?

I didn't really let bags sit out more than a day and I never pre sealed them either. If I couldn't inoculate I would just leave them in the pc until I was ready. I just sanitized my gloves, not the bags.
I had a lot of pc's (fifty 5lb bags per run)
I had 3 x 941, 2 x 930, 3 presto 23quart, and a 75x

It wasn't my house, and the landlord saw my setup. He was already charging me more than he should for running my 75x and electric heat.
You can't see it from the pic but I had french doors that were open and a 24" industrial fan exhausting. And a window open on the opposite end, so the airflow was quite good. I had it under control and someone was always there with a fire extinguisher.

Can I get your guys opinion on this concept. I got a lot of reddit hate (like a lot) from noobs when I suggested it.
https://www.shroomery.org/forums/showflat.php/Number/27506021

TL;DR its starting cultures by using mini grain jars to get myc started, then transferring a colonized grain to agar.
The argument is why not just start with a drop to agar, its just an extra step and another chance of contamination.
My argument is that i previously used these same syringes to agar and got nothing but bacteria after waiting 3 weeks. dishes pictured in the thread are 15 days from inoculation of grain. 10 days after inoculation the transfer was put to agar and it grew out for 5 days.

if it works it works but why take the syringes to grain if they were already bacterial on the plates?  u want to try to get some germination and then clean it up after you put it to agar?

yeah, unfortunately I can't keep them in front of the flowhood - i just stack them on shelves  until I have like 100. Before using them I  wipe the top  of the bags with iso  then  cut them open.
I spend  quite a lot  of time in front  of the flowhood and my  success rate is good but the amount  of iso fume i breathe makes me  gag every time  and just made me  question whether  iso is even doing anything.

Next time I will try a 10% bleach solution to spray and wipe bags on 1 round of bags, and a soap + water on the other.

I am not  trying to cut corners here I just think we are religiously using too much iso:D

i was waiting 3 weeks and not getting germination, just bacteria. For me this worked faster and I got clean plates after the grain transfer

what are the reasons to seal bags when using a steamer? Does a steamer not cool down fast enough to make a vacuum seal?

it makes a seal but it takes a very long time to cool down and could possibly introduce contams.  maybe u can get away with not doing it but its what I do.  plus I cant fit 50 bags in front of my hood at once so it gives me extra peace of mind knowing that they're fine sitting out of the flow

I tried bucket tek again, and I think I like it. Definitely limited by the water heater it seems 4 or 5 really good ones back to back, but sunce it's prep work I can just do sets of 4 and stack em up. Got 10 to spawn now and I'll prep the next 10 today. Getting caught up

I posted this in that thread too:

When you pull colonized grains to agar you'll get much faster growth due to the grain nutes you're bringing along.  If you do a transfer to just agar after that they'll slow down considerably.

But I like the syringe => grain => agar route for germination and culture isolation - see https://www.shroomery.org/forums/showflat.php/Number/26369976/fpart/all/vc/1 for a whole PE isolation grow log using this method. :awesomenod:
