I picked up a fresh pack of nitrile gloves to do clean work. Seems like my no glove work has been sloppy. Damn things went up like 100% since COVID. Mostly just satelites at plate edges, but I think that's half my problem at this point

I don't use gloves, nor a couple people I know. Ide almost be worried about something else persobally. I mean unless your isoing your gloves after like every movement they won't be much dirtier than isod hands. Me at least I'll move in and out of the flow consistently.

It's the edges of the plates and takes over a week to be noticeable. I just replaced the A/C setup so there will now also be less cross current near the flow. Just figured I'd cover my bases ya know.

Plates are proofed I'm sure. Your wrapping them in front of the flow obviously, with it on. I have a fan that sometimes get left on when I'm working, hasn't effected me that I've noticed yet.. as long as your flow is stronger than your ac, and esspecially if your in the front 10" it shouldn't matter I would think. The air shouldn't fight unless maybe your ac was really pushing towards it. And I feel ya, just trying to spit ball with you.
I'm going to do an iso experiment soon testing sanitized steal to agar. I also wanted to test a sanitized glove and finger. I've considered gloveless hands a bunch, and idk

AC currents and fans are totally a big thing to consider as well as door location when placing your hood. Simply opening a door next to a hood will create a huge rush of air. The breeze of the hood isn't that strong, even breathing heavy could overcome it.

I just figured since air flows the path of least resistance if it's less than that of the flow it wouldn't be a problem. I don't open doors when we're working unless it's necessary, but never work when the door is being opened or closed usually. But I have noticed the hood I made compared to my ffu has drastically different rates, but neither effect performance in my case. The one I made feels twice as heavy but idk.

my tam rate plummeted since i got the FFU. I try to do all my work now right up near the filter. the SAB was a fucking nightmare.
I left a plate open in the flow for 8.hours onthe 17th and it's still pristine, tho maybe it dried out too much to show tams
also a plate i labeled and didn't nocc from the same day.

I agree. I'm constantly wiping my gloved hands and arms with iso when working. I would have horribly dry skin if I didn't wear gloves which is my main reason for wearing them.
the main thing to remember is to iso your work beer

I dnt see the point in using iso on your gloves and arms. I mean, if you aint rubbing at it for 15-30sec, there'll be no sanitization. Use lysol wipes, people!

I actually use a liquid sanitizer. It's 80% alc with hydrogen peroxide and propolyene glycol. It lingers long enough.

Btw, the hydrogen peroxide in hand sanitizer is supposed to be degraded by the time you use it. The purpose is to kill any spores in the sanitizer itself, ensuring sterility. Low dose H2O2 works much too slowly to act as a sanitizer in its own right.

Hydrogen peroxide: used to inactivate contaminating bacterial spores in the solution and is not an active substance for hand antisepsis.

yes, rubbing them with an iso soaked rag. not spraying my self and rubing in with my hands. if your working in the flow for extended periods of time and stop to take sips of beer every so often its not a bad precaution to take.
lysol smells like shit compared to iso IMO and id rather have iso on my skin than lysol. not to mention it costs way more

That could be it, the h202 or PG leaving residues eve , or the high alc content... I heard 70% is best because of osmosis. To go along with this sanitation experiment I also testing sanitized steel, sanitized fingers and gloves, but also with 70%vs 91. I couldn't find 99 when I was at the store. I do have some everclear tho

I constantly iso my gloves too, especially if it’s something important or especially vulnerable like lc.
Sometimes if I only have one or two plates to do I’ll just spray my hands down but it’s so dry here as it is that’s not a great method.

no ones doing the hydrogen peroxide followed by vinegar method?
I use iso on my hands and shit too, but for the work space I've been doing the above combo ( one at at time, not mixed)

I just wipe everything with iso paper towels
I had some poured petris out for months and they were clean where they were but I moved them to my old grow closet(where I grew lots of mold) and all my old clean plates instantly molded
Think I'll clean that closet

#3 from here good read

Frequently disinfect gloved hands with 70% ethanol while doing aseptic work.
Although the gloves may initially have been sterile when first worn, they will no doubt have contacted many nonsterile items while in use. ..
