Spore print to liquid culture?
    #27469455 - 09/15/21 02:24 PM (3 years, 4 months ago)

Ok so I have made around 20 spore prints from a really good strain I like.  Is it possible to just scrape a bunch of spores into the LC and go from there?  Or do oi need to make a syringe first?

Thanks

I think that’s what syringes are right

You can do it but chances of success are very low in my experience. Spores aren't always clean and scraping things into LC provides a lot of opportunities for contamination. Best is to put spores on agar, do at least 1 transfer to see clean growth, then put a small piece of colonized agar into your LC jar using sterile technique.

No. Don't go spore to LC. It's stupid. Sorry for sounding rude but it is. Spores to agar, agar to LC. Even then it can be fucked up easily. Spores to LC is a fail even if you succeed.

Edit:  Got beat to it again.

This.

I did spores in Syringes, packaged as good as you can get, to LC for years! 99% failure rate. Spawning grain to monotubs with Liquid culture being the inoculant of the grain jars, ALWAYS ended in GREEN.

I  can't tell you how many times I tried this. In hind sight it taught me never to try the same thing expecting different results. After I did Agar wedge > Grain > Mono or shoe boxes, I ALWAYS got mushrooms. Even if it contaminated on the first, second or third flushes.

My goal was to get clean spawn that could flush until the cake was spent. A shriveled up hunk of mycelium barely resembling the huge fluffy cake it originally was. I never got to completely clean spawn though. Looking back I think my agar plates themselves were a source or contamination because after the first inoculation of a plate I would store it in open air with parafilm. Handling it with unclean or semi clean hands. I never wiped down the plates before make new transfers or inoculating a grain jar. At some point I will pick it up again and those are some of my goals; clean culture/spawn.
