I read your Tek, but it sounded like you use it straight away / don't store it or grow it out?
I was planning to use it with speedy culture and fully colonized plates.
Does your approach work with wedges or 1/4 plates?
...Also, I went to all the fuckery of making lids with holes in them just for LC :p

thats what i read in the josex poke thread.
i made these like 10 days ago for fun just to try it. I've stopped shaking it like 4 days ago. It's filtered brf lc with agar.
Made them with the intention of one and done. If I opened them I'd just use em all at once.

I just use media bottles and I tighten em down to shake. Maybe that's why my LC are slow to colonize, but I also don't swirl em enough. Took a day off to help someone move yesterday and now I'm pretty beat.

Oh, I meant if you splash the filter it might start growing mycelium on it and could be an issue, if your not going to be using it soon…. My bad I should of explained better.

I'm gonna abandon the modded lids and give the baby bottle idea a go. I'll report back progress
Wide mouth, heat safe, easy to pour, cheaper than a media bottle. Awesome idea 😎

The foil lids work but you have to take them off once to inoculate with agar and they are fragile. Easy to mess it up accidentally. I was using o-rings I had that just happened to fit snug on the neck to kinda seal them but you could use rubber bands after PCing and transferring or probably nothing.

I wouldn't assume that, pc some in a foil pack to test it.
The problem is the inside liner of the lid that actually is the seal melts even if the lid itself is OK.

usually they use this blue colored silicon liner. isnt silicon really high temp resistant? my sparkling water lids are like that too and when i shake it nothing leaks. if plastic mason jar lids work i dont see why this wont.

I just tested the lid with a soldering iron that has digital temp control and it totally melts like butter at 250f.
That liner on water bottles is definitely not high temp I promise.

Yes, I do know that, but it's irrelevant to my question. I want to know how to plan for long-term storage of varieties that I don't plan on growing every year. If parameters like refrigeration and air-tight containment help increase the half-life of spores from, say, 2 years to 4 years, then you bet I will keep them like that.

When I looked into this, it seems cool stable temp, dry, no air flow, and dark.
Your home fridge is probably bad as temp fluctuation likely worse than a cool dark cupboard in a sealed container.
If you want reliably longer than a few years, there's a research paper that said longest preservation of cultures was achieved by refrigerating myc in distilled water (it goes dormant if prepared properly). I think 14yrs in the paper was the longest.. I could be wrong about that number, but it was a long time.
