Why are there more than one sector? And what does a monoculture do when u finally achieve one? Still have never taken a clone. Very interested.

more than one sector could indicate different strains. a monoculture will express the genes of a single strain. that could be good or it could be not so good. if you have say a clone plate and you make a transfer to a new plate and see a tiny bit of sectoring but you take it to grain and it kicks ass then I wouldn't worry about isolating it further. if it sucked tho I wouldnt attribute that to there being different sectors. in that case I would just go back to ms and look for a new clone candidate to take and test

Yeah most clones show some sectoring ime
Not necessarily a bad thing though, most people dont really need a true isolate
Sometimes clones (or even ms) with multiple strains present will be just as organized as an isolate, to a point where its hard to distinguish it from an isolate.
I have isolated a few cool strains from clone sectors though, but its not really useful for anything but strain hunting

as far as i understand, might even be problematic to separate them
i really wish i knew more about the dynamics of multistrain vs isolated strains. the way strains interact in a colony is pretty fascinating but i dont even know where to begin to learn more about it, besides experimentation
if anyone knows any good resources on this topic, for any mushrooms not just actives, please share

I'm just about to experiment with this myself. I have taken 7 different transfers from one clone plate and I'll see how the results differ.

you gotta have your variables VERY standardized and run everything with lots of redundancy to even be able to tell, its kinda a pain in the ass
id say at least 3 of each thing you are testing, ideally way more
if you have something that performs well, it really isnt worth the effort unless you just enjoy strain hunting
the VAST majority of deviation in grows is more to do with environmental/prep factors than genetics. this is why even when working with the exact same genetics you will see vastly different results from different growers (in terms of fruit morphology, yields, canopies, etc). even when its the same grower subtle differences can make a huge difference. standardizing things to a point where you can get meaningful data on genetics is pretty difficult

I think he self bans on a regular basis.
Do people think sectoring is bad?? I’ve never gotten that from anything I’ve seen here.
I just think of each good culture like a voltron. If you remove a sector it’s like cutting a leg off.
Now if you’ve got a culture that needs some work removing a sector could be like cutting a huge weight off and now it can really move.

Well I hope he's back soon. I'm going to make some petris in actually petris, not Togo cups, so I can test sanitized items on it. Also im going to test some containers, like Togo cups, coke bags, ziplocks. Rather than pouring directly in them tho I'll take transfers from petris and put them in to different containers to test their sterility.
I've also wondered that aka, was going to ask the other day if if isolate is preferable. I wouldn't see any reason so except maybe on terms of vigor, maybe an isolate would grow fast because it all "working together" idk.
But I've had some culture I could keep transferring and they always seem to sector a little...

Has anyone adding agar to LC noticed anything about identifying contams? Like, it makes it hard to see if it's turbid... Also im wondering if I can expect to see present molds or bacteria growing on the surface if there is a contam?

Isn't lc impossible to know until you test on agar? (Besides really obvious things like green growth)
Anyone know why APE get all soft? Is it just ripening/rotting? Does it lose potency or something?
