Yeah the syringe is poly stuffed so I can blow up the bag after pc’ing via fish tank bubbler.

I vac seal the swabs because I open the bag end submerged in H2o2 (in a modified glad container) and then slide the swabs in through the opening (think of a beavers den opening) so the inside of the bag doesn’t get exposed to room air.. then I seal the bag end back up while it’s still submerged.  There’s a small set of SS scissors that’s pc’ed in the bag that I use to cut the swabs open.

Where's the hand drawn schematics? Those are legendary.

  here’s the first part of my cloning procedure (the serial sterile water rinse) using the oven bag.. maybe when I finish that one, I’ll do the swab to spore water one lol.. they take a minute

Seriously guys! Look at this fucking artwork.

Woke up to probably last outdoor flush of cubes for the season. Don't like cubes and decided to try some pans :thumbup:. Should I nightcap these guys? Guessing ~10g fresh.

I have to agree. I can understand tilting the master jar so that the handle does not hover over the mouth of the jar. Sound method imo. But not sterilizing the spoon beforehand and only sanitizing with 70% iso sounds like a disaster waiting to happen. Definitely bad info for any noobs that may end up reading that.

i did an experiment today. i have to work with a lid under my sab cause the counter i do my work on is too narrow, but it definitely causes extra shaking and vibration and my armholes are too small so i bump it way too much. so i took an agar plate and opened it, then bumped the sab a couple times. i wonder if things will grow or not.

sterilize that spoon in a jar and open it in your sab at the least.

LOL look at my recent history, you may be fucked!
I went from

To

All because last time I cleaned the SAB I put my lid back on the bottom. I'm curious though maybe I just have a crazy mold load.

i do have plates that kind of make no sense, like i took transfers from clean portions, and the next transfer is all wacky, but i make a lot of transfers so i get enough good plates to work with. i wonder how much of it is bumping the sab, but i had a lot of contam vectors i had to work on recently so who knows.

i wish i had more counter space cause my sab wont fit without the lid. im in a closet working (i crack the door so i dont get a vacuum). one thing i do though, is ozone the closet for 20min before i work. so i wonder if even there are particles stuck on the sab, theyre all dead because the ozone. im really curious to see what happens to that plate.

I would seriously discourage adopting non sterile techniques in place of sterile.
Do not spoon sterile grains with non sterile instruments.

Do you see any difference between scooping out grains with iso soaked fingers and spoons... Because it's exactly the same thing, don't do it.

Exactly.  Although there is quite a difference between skin and stainless steel.  When i origionally commented on mr mushes post it was because i doubted that the scooping was the cause of contams.  It can and does work if done properly but its not the best method and I dont use it anymore.  I was experimenting when i did it and even though i had great succes i found better methods and would not recommend anyone scoop over pouring.  It takes much more care and was not a time saver in the end so it got ditched

Is the idea of a scoop because you're doing bag to bag g2g? Pouring from a jar seems easier than using any sort of scoop? Like, it's dead simple and fast.

If youre growibg commercial jars would be a PITA.  So scoop from bag to bag would be better but its easy enough to pour.  The main reason i can think to scoop over pour is a more acurate volume measurement but whats the point

I saw Myer's using a SS scoop in one of his videos but it might've just been scooping spawn in bags of substrate IIRC, not sure if that requires perfect sterility but I did think huh how does he get away with that.

I think it's also probably a little different if you have somewhere guaranteed sterile you can place the scoop between bags and you're working in front a wall of flowhoods.

I'm also str8 up bought to order one of those red plastic scoops I've been using a plastic pint container I need to stop half assing it with the tools

If your competent in technique a scoop is fine but I wouldnt recommend it to someone that is still figuring out what works best.

But everytime you touch the end of that spoon and put it back into the jar you are increasing the chance of introducing mold or bacteria. Just my humble opinion. You do you, brotha.

Dont knock it till you try it.

I wouldnt do g2g like that tho... just sawdust ha

How much worse is it than lifting thag bag over your other bag? And idk about you giys but your hands stay very unsterile why in the lab, unless your sticking your fingers individually in your bactizapper ha.
But we mix bags, move bags, grab another one and then go back to out master inoculum and pick ot up, and put it directly over our substrate. Nothing sterile about that.
I couod understand sterilizing the spoodle in a pc before, but still havent found it necessary.

So there is a benefit to scooping!

Ive found for my bag/hood size it is easier and a time saver.

And you had great success....

Whats your bettwr mwthid unless its pouring? Cause spoodle is easier for me by far. I use 10lb spawn bags an 12lb sub bags. Can be liftin in my 2x2 flow.

I cant believe so many people are like no it doesnt work, when it clearly does.
Dont knock it til you try it.

I think I might’ve done something weird to my cultures by keeping them in extreme heat.

It’s been probably 100 average in there the last few months and for sure hasn’t gotten below 80.

All the plates grow fine for the most part, but I haven’t been growing because it’s so hot. A week ago today I finally knocked up 8 jars. Only 2 have jumped off, the rest look like I dropped the wedges ten minutes ago.

This is a taz clone that was extremely quick on plates but hasn’t moved in a week.

Hopefully they wake the fuck up soon.

I’m kind of wondering if since they’ve never been in cool temperatures before if the move from the 100 lab to the 73 grow room was enough to make them hibernate for now.
