Hey guys of all genders,
So this is my Josex' Poke LC from way back. Been colonized and been swirling. Wanted to nocc millet with it later today.
Is it looking funky? What's going on in there. I don't know if I can test it on agar cause once it's open I figure byebye, right? It's okay if oats fuck up but FFS millet is $2/lb!

Is that just pinning in there?!? Or is it trash? I also have a wedge drop LC in a media bottle. Can I aspirate such to test on agar and be able to use it again?

Space, to my eye nothing indicates that the first LC is bad, but we can't be sure from a pic. Looks like a healthy, thick LC that's a bit grown out.
The others are just way too dark to make out anything, pics are usually terrible to judge cleanliness of an LC.

Yeah, I know...the first is your poke into specifically prepped broth.
The other two... ...agar prep that I forgot the agar for.
I'm just goona throw caution, and maybe $$$, to the winds and nocc 10 millet with the 150 ml of thick JoPo LC.

Just don't wim Hof hold for too long.
The darker LC's do look suspect though, but it could be you just used way too much nutes to make the broth so it's very dark and cloudy.

Haven't been at it for a while. Have medical O2 which I used for WH breathing once and got 2 SIX MINUTE breath holds. Cool AF. Felt like a mutant. lol

I think i will poke this T2 in 2-3 days for my first try doing this method, that satellite you see is just agar peace from transfer, wish me luck i was waiting to long to try this

I never looked up on how to make LC and the very first thing I found, when I finally did, was your tek. I got everything I need and will give it a try!
Edit: I decided to do the grain broth you like so much. I just got a question regarding the LC jar. I got these wide mouth mason jars with a metal lid a a ring to screw it on tightly. Now when I want to sterilize the jar with the broth, will I have to flip the lid and screw on the ring tightly or loose, in order to avoid pressure build up? Or can I just use one of my modified jars I use for my grain spawn (metal lid with hole covered with micropore tape for GE)?

Some people use unmodified lids for LC with success but I don't recommend it because you have to take certain precautions which are a bit of a nuisance that could be entirely avoided with the use of a filter.
I barely trust mp tape for anything and even less for LC. It's gonna get pretty wet and it's not a good filter to begin with. SDF's (whole or cut out) will work, as well as properly applied/stuffed polyfil and of course, a watman filter that can be purchased cheaply on the internet.

I take it your lids are not designed for aspirating into syringes? In that case you can use an inoculation loop to do the job.
Flame the inoc loop, crack the LC lid and quickly reach with the inoc loop into the LC, then streak on agar.
Sometimes with this method you won't see any growth on agar at all because it's possible you didn't catch any mycelium with the inoc loop. But even when no myc takes on the agar, if no bacteria grew either you can confidently say the LC is clean, because if there was bacteria it would show quickly regardless if there was myc growth or not.
Eventually, if you plan on sticking with heavy LC use, the goal is become good enough at it that you trust yourself and don't even feel the need to test them.
I never test them myself, the few times I did get a bad LC I could tell with ease just looking at it and I'm willing to take the chances (directly going to grain) if for whatever reason that one-off bad LC escapes my visual inspection, but I can't remember that happening besides that time that I got a moldy LC that got the OK from me. Bacteria in LC is usually much easier to spot than mold, though.

The turbid looking ones are w/ agar. JoPoked nocced the broths on 8/2. I expect them to really explode in this coming week.

Dual comparison of transfers! With such a huge area to work in, if your technique is bad your see exactly which way your arm came over the huge sandwich box.
I should say left hand of box was a blade, right hand was "poked"

I know what that feels. My mushcult shed recently got flooded with heavy rains and contaminant spores are everywhere despite working in a SAB armed with intermediate techniques.
